Morchella is a kind of high nutrition and low caloric nourishing food, both thefruit body and mycelium contain rich nutrition composition. Along with theunceasing raising of people's living standard, the demand for Morchella increasescontinuously. The burliness course and mechanism of Morchella have not beenunderstood so far, the conclusion that a lot of experts in artificial culture reachedcan not be effective reliable repeated and verified, therefore industrializationproduction of Morchella fruit body has not succeeded. The production ofMorchella is relatively easy.In this paper, we tried to add trace element,Germanium in culture medium, and made Morchella enrich organic Germanium;and improved the spectrophotomety for surveying trace Germanium, made it moreaccurately measure the Germanium of Morchella; besides we studied the dry wayof Morchella and the prescription of Germanium-enriched Morchella tea of healthprotection, we find a new way for making use of Morchella.1. Confirmability of the research content. We summarized and analyzed the related research for Morchella, pointed out the mechanism that using edible fungi to enrich trace element, advanced the assume that using submerged-fermentation to make Morchella enrich trace Germanium, offered theory basis for the comprehensive use of Morchella mycelium.2. Improvement of Germanium measurement. On the foundation of forefathers work, improved the measurement of trace Germanium in Morchella mycelium, show by research: selected wavelength was 497nm, its standard curve was GA = 0.1957GC + 0.0247 , its correlation coefficient R = 0.9725. When we didn't adjust pH-value of the cineration things, cineration temperature was 600℃and cineration time was 1h, the survey result was relatively ideal. Now, the rate of test recovery was 87.15%, accuracy was 12.85%, precision was 0.89%, method limit of detection was 0.0002mg/L.3. Determineation for the extraction method of polysaccharide. We adopted stepwise extraction to extract polysaccharide of Morchella, had studied the factor that influence extraction rate(extraction temperature, extraction time,extraction proportion), knew by variance analysis that the order of primary and secondary of factors was: extraction temperature, extraction time, extraction proportion. The best extraction technology factors were that extraction temperature was 98℃, extraction time was 120mins, extraction proportion was 1:20, then the extraction rate of polysaccharide was 5.1882%. Compared microwave handling with supersonic handling, the result showed that microwave handling was better than supersonic handling. When taking off albumen of polysaccharide, it was taking extraction rate as index, volume proportion of chloroform and butazolidin, volume proportion of Sevag reagent and sample and oscillate time as factor, carries out orthogonality experiment. Knew by variance analysis that the order of primary and secondary of factors were that volume proportion of chloroform and butazolidin, volume proportion of Sevag reagent and sample and oscillate time. The best technology is that volume proportion of chloroform and butazolidin was 4:1, volume proportion of Sevag reagent and sample was 2:1, oscillate time was 30mins.4. Optimization of the culture condition. Through testing we chose wheat bran and soybean powder for the culture medium, most suitable ferment period was 5 days, the influence of frequency of shaking table and temperature for mycelium biomass was more obvious, in this test we chose 120rpm and 28℃. Germanium influence for the growth of Morchella mycelium was serious, low density of Germanium had promote the growth for Morchella, but when Germanium density exceed 100mg/L, with the increasing of Germanium in the culture medium, mycelium biomass reduces gradually. Useing twice orthogonality combination test, after handling test data, we got the optimization equation: y? = ?49.9304 +1.0070 Z1 + 0.6239 Z2 + 0.0146 Z3 ? 0.0084 Z1Z2 ? 0.0016 Z2 ? 0.0004 Z3 2 2. Through calculation, we got that when Germanium density was 18mg/L, temperature was 26℃and frequency of shaking table was 127rpm were the best culture condition of Germanium-enriched Morchella.5. Research of the growth curve. Regress with the modified Gomperta equation for Morchella growth curve equation, regression effect was drafted better and had very high level. Its correlation coeffient R=0.9759. We got the growth curve equation of Mrochella: y = 0.8564exp[?exp(1.7173?0.4047t)], its related index R=0.9759, now the prolong period λ= 1.7857h , the biggest growth speed μm = 0.1266g / h ,...
