CHAPTER 1 In this chapter, the fundamental contents of supramolecule, dye, cyclodextrin and protein were simply introduced. In recent years, the studying progress of the interactions of dye with cyclodextrin, the interaction mechanism of organic dyes with proteins and several analytical methods of BSA analysis were reviewed in details.CHAPTER 2 In this chapter, a polarographic method for thedetermination of acid scarlet GR(AS GR, C.I. Acid Red 73) has been established .A stable and sensitive linear-sweep voltammetric peak has been obtained at -0.85V(vs.SCE) in NH3-NH4Cl (pH 8.84) buffer solution. The peak current is proportional to the concentration over the range of 1.0 × 10-8 mol·l-13.0×10-5 mol·l-1 (r=0.98840.9994) and the limit of detection is 9.0 × 10<sup>-9 mol·l-1.The relative standard deviation wasl.90%(n=8) and the mean recovery of acid scarlet GR was 99.60%103.5%. The interference was determined about more than ten kinds of dyestuffs. This method was anticipated to use in the quantitative analysis of acid scarlet GR. At last the mechanism of electrode reaction was preliminarily discussed. CHAPTER 3 In this chapter, the supramolecular systems of acid scarlet GR with cyclodextrins have been studied by polarography. The result shows that acid scarlet GR has formed 1 '. 1 inclusion with diverse cyclodextrins. The inclusion constants were calculated, and the inclusion ability of diversecyclodextrins was compared, which provided some data for further application of acid scarlet GR and cyclodextrins. CHAPTER 4 In this chapter, a polarographic method for thedetermination of acid cyanine 5R (AC 5R,C.I. Acid Blue 113, CI: 26360) has been established and the supramolecular system of acid cyanine 5R with cyclodextrins has been studied by polarography. The result shows that acid cyanine 5R has formed 1 '. 1 inclusion with diverse cyclodextrins. The inclusion constants were calculated by "electric current method". Furthermore, the inclusion ability of diverse cyclodextrins was compared, which provided some data for further application of acid cyanine 5R and cyclodextrins. CHAPTER 5 A supramolecular complex of bovine serum albumin (BSA) with acid cyanine 5R has been shown to form in Tris-HCl buffer solution (pH7.42) by linear sweep voltammetry (LSV), fluorimetry and spectrophotometry. The binding ratio and binding constant of BSA with AC 5R have been detected by LSV and fluorimetry. The binding mechanism is also preliminarily discussed.In Tris-HCl buffer solution (pH7.42), AC 5R can easily be reduced on the mercury electrode and it has a well-defmed LSV peak current (ip) and peak potential (Ep) at -0.65V(vs.SCE). In the presence of BSA, the peak current (ip) of AC 5R decreases and the peak potential (Ep) shifts to a morepositive potential. The decrease of the second order derivative or reductive peak current ( A ip" ) of AC 5R is proportional to logarithm of BSA concentration in the range of 1.54 X 108moH'11.54 X lO"3 molT1 (r — 0.993 l0.9977).The limit of detection (LOD) of BSA is 9.0 X 109 mol-1'1. The relative standard deviation (RSD) is 1.83%(n=10) and the standard recovery is 97.5%—'104.8%. This method can be used to determine BSA concentration based on the interaction of BSA with AC 5R.CHAPTER 6 In this chapter, the supramolecular system of bovine serum albumin (BSA) with acid scarlet GR has been proved to form in tris-HCl buffer solution (pH 7.43) by polarography. The binding ratio(l :1) and binding constant 0 =1.167 X 105mol"'-l) of BSA with AS GR have been detected. The property of polarographic current has been studied. Furthermore, the binding mechanism is also preliminarily discussed.In tris-HCl buffer solution (pH 7.43), AS GR was used as a good electrochemical probe to determine the BSA concentration in aqueous solution. The second order derivative of polarographic reductive peak current (ip " ) of AS GR is proportional inversely to BSA concentration in the range of 3.0X 10"6 mol-r'-l.OX 10'Vol-r1 (r=0.99060.9953). At the same time, the spiked effect has yielded in the solution of AS GR with BSA of micro concentration, which makes the ip " of AS GR is more two times than before and it is proportional directly to BSA concentration in the range of 1.0 X 107 moH"l3.0X lO^moH*1 (r=0.9919-0.9987). The current response of higher sensitivity is of significance to the determination of micro BSA.
