Benzimidazole and its derivatives possess a variety of biological characteristics, widely used as Pharmaceuticals, Veterinary, Anthelmintics, and Fungicides, et al. Metal derivatives of benzimidazoles also can be used as analogs of natural moleculars.In this paper, 4-fluoro-6-methylbenzimidazole is prepared from 2-fluoro-4-methylaniline by acetylation, nitration, hydrolysis, reduction, cyclization. Factors influencing the product yield are discussed, such as nitrating agents, reductive agents, reaction temperature, reaction time, have been investigated.2-trifluoromethylbenzimidazole, 2-methylbenzimidazole and 2-octylbenzimid-azole are synthesized by the condensation of o-phenylenediamine with trifluoroacetic acid, acetic acid or nonanoic acid respectively.1-(p-methoxybenzyl)-2-(p-methoxyphenyl)-benzimidazole and 1 -(p-chlorobenz-yl)-2-(p-chlorophenyl)-benzimidazole are synthesized by the condensation of o-phenylenediamine with p-methoxybenzaldehyde or p-chlorobenzaldehyde. And there is no report about the synthesis of 1-(p-chlorobenzyl)-2-(p-chlorophenyl) -benzimidazole.5-nitryl-2-trifluoromethylbenzimidazole is prepared from 2-trifluoromethylbenzimidazole by nitration of 2-trifluoromethylbenzimidazole.
