The objective of thesis was to develop a fermentation bioprocess by a strain of Armillariella tabescens and to study the extration and purification of Armillariella polysaccharide and the effect in pharmacology. The research content included, (l)the technology of solid fermentation and liquid fermentation condition of Armillariella; (2)the extration and purification of Armillariella polysaccharide and simple configuration identification; (3)the effect of Armillariella polysaccharide in pharmacology. Main results are as follows:1. The research result of solid fermentation: Confirming the optimum time is 45d according to the content of polysaccharide. Several enzymes were determined in the process of solid fermentation, which can make us master the metabolism rule and growth state of Armillariella. This fermentation system has some advantages, such as long-Cycle, which make for the synthesize of officinal ingredients. But this advantage also is a flaw, which restrict the industrialization production.2. The technology of optimizing disposition of system factors were adopted to optimize the liquid fermentation conditions of Armillariella tabescens(Scop.ex)Sing. The result showed that the yield of dry cell has reached 3.83g/100ml which was increased by 65% compared with the best result ever reported .In addition, the studies about the growth of the strain and the metabolizing of products rules were showed in details, which provide academicbasis for further industrialization production.3. The results of single factor test and orthogonal test proved that the optimum extracting conditions were as follows : the ratio of sample to water ( w : v ), the extraction temperature and the extraction time were 1:5, 90°C, 2.5h respectively. Through decoloring, removing protein, cellulose chromatography, the relative pure Armillariella intracellular polysaccharide can be got. Two factions are found by electrophresis in Armillariella intracellular polysaccharide. Through IR and determining sulfate positions, it can be tested that there are hydroxy, sulfate positions in Armillariella intracellular polysaccharide4. The effect of Armillariella polysaccharide on mice immunological function was proved by means of determining the production of the hemolysin antibody, the activity of NK cell, the function of macrophage and the production of IL-2. At the same time the result can be tested that Armillariella polysaccharide can not only protect the mice' liver but also inhibit the tumor in mice through detecting ALT in serum of mice and IL-2 and NK cell activity of the mice with tumor. Those research results can indicate that Armillariella polysaccharide is an available and valuable substance.
