Armillariella tabescens, which can be named bright bacteria, is a large edible and medicinal fungus. Because the mycelium could produce blue fluorescence in the dark, we call it bright bacteria. According to the related literature reports, this fungus has certain curative effect on hepatitis, cholecystitis, tumor and leukopenia caused by chemoradiotherapy, and it also has certain health care efficacy. Armillariella Oral Solution is a traditional chinese herbal pieces which contains armillarisin A, polysaccharide, amino acid, polypeptide and varieties of bioactive substances.At present,the Armillariella tabescens preparation was generally carried on with solid state fermentation. The solid medium was made of corn flour and potato powder which according to certain proportion of them. The advantage of this manufacturing technique:make for a variety of active ingredients of medicinal expression and accumulation; The disadvantage of this manufacturing technique:had a long production cycle. Therefore, the solid state fermentation restricted the production efficiency of enterprise such as HeFei Cheng Zhi Biological Pharmaceutical Co Ltd. With the increase of culture generations, The Amillariella tabescens degenerate and cut down the production and drug efficacy. As a result, we hope to find a way to rejuvenate the degenerated Armillariella tabescens.Our laboratory has carried out a series of research. Specific content are as follows:(1)The study for application of liquid strain and the optimization of liquid seed culture of Armillariella tabescens;(2) The preliminary research of liquid fermentation of Armillariella tabescens;(3)The methods optimization of extraction of Armillariella polysaccharides;(4) The technology improvement of solid state fermentation of Armillariella tabescens.On the basis of related literature reports and preliminary work, It was discussed in this paper about the optimization research of fermentation technology of Armillariella tabescens with taking the deep liquid fermentation in fermentor and stationary liquid fermentation methods, and analyze the feasibility of taking the place of traditional solid fermentation. At last, we carry on the preliminary study of rejuvenation of degenerated Armillariella tabescens.The main research content of this paper:(1)The technology research of deep liquid fermentation of liquid seeds;(2)The technology research of stationary liquid fermentation of liquid seeds;(3)The analysis of components in fermentation liquor;(4) The preliminary rejuvenation study of degenerated Armillariella tabescens. The results are as follows:(1) The research results of deep liquid fermentation of liquid seeds in fermentor: The deep liquid fermentation adopt the measures of28℃, ventilation1:1v/v·m,200r/min, culture7days, the biomass of Armillariella tabescens in dry weight was16.55g/L, and the mycelia contain polysaccharide5.42%, protein1.75%. The results show that the deep liquid fermentation of liquid seeds in fermentor could get a high biomass in a short production cycle.(2) The research results of stationary liquid fermentation of liquid seeds:The stationary liquid fermentation use500mL conical flask to shake3days in the condition of28℃,150r/min, then stationary culture14days, the biomass of Armillariella tabescens in dry weight could reach to10.69g/L, and the fermentation broth included polysaccharide1.016g/L, protein0.320g/L. the surface mycelium of fermentation broth secrete large amounts of brown liquid, and the strong and multiple branch shoestrings were full of the fermentation broth. The results show that the technology of stationary liquid fermentation is superior to the traditional solid state fermentation.(3) The analysis results of components in fermentation liquor:With assurance of biomass and active ingredients,this two fermentation methods are greatly shorten the fermentation period, and both superior to traditional solid fermentation, and could use for reference to the industrialization production of Armillariella tabescens preparation. Although the stationary liquid fermentation was inferior to the deep liquid fermentation in the biomass, the former had a deeper fermenting degrees. To the further study of stationary liquid fermentation, we found that the content of armillarisin A in the fermented liquid could reach to3.118mg/L. (4) The preliminary research results of making degenerated Armillariella tabescens rejuvenation:We explored the rejuvenation effect of degenerated Armillariella tabescens by adding different concentration of willow wood chips extract, straw extract and different kinds of trace elements to the ordinary PDA solid medium. Preliminary study show that it was obviously rejuvenative when we added different concentration of willow wood chips extract into PDA solid medium. To the further study of adding concentration of straw extract, we found that1%concentration of straw extract was the best adding concentration.The stationary liquid fermentation is better than the deep liquid fermentation, the former fermentation technology could not only cost saving, but also easy to operate. It Can take the place of the traditional technology of solid-state fermentation; we also researched the way to make degenerated Armillariella tabescens rejuvenation. These studies provided theoretical support and reference data for HeFei Cheng Zhi Biological Pharmaceutical Co Ltd to product Armillariella Oral Solution, it has certain practical significance and application value.
