By comparison of six different Coriolus versicolor strains, strain LS was selected for polys-accharopeptide production by submerged fermentation. After optimization of shake-flask culture,polysaccharopeptide production was scale-up to 30 liter fermentor. Maxium biomass andpolysaccharopeptide yield were 24.29g/L and 1.63g/L respectively. Total sugar content was 50.41%,peptide content was 35.77%, reducing sugar was 0.53%. Its HPGFC Chromatogram was consistentwith that of standard. These indexes were in accordance with China medicine standard WS-XG-021-2002.The culture medium was further optimized. The medium composition was: 8% soluble starch,4% bean cake flour, 0.2% corn steep flour, 0.5% wheat bran, KH2PO40.15%, MgSO40.02%. Thesuitable shaking culturing process was: the initial pH of medium is 5.0, culture temperature is 26℃,volume of medium is 50ml medium in size 250ml shaking flask and inoculum size is 10%. WhenCoriolus versicolor strain LS was cultured 6 days on these conditions, the maxium biomass was35.001g/L and polysaccharopeptide yield was raised from 0.718 g/L to 1.726g/L. Extraction condition of polysaccharopeptide was studied. When using homogenizationtreatment, the optimal extraction condition was: extraction temperature 80℃, adding water volume6 times, extraction time 2 hours, extraction times 2 times, the voluminal ratio of ethanol tosupernate 3 times and deposition temperature 4℃. Under these conditions, extracting rate ofpolysaccharopeptide was 66.86mg/g. In addition, different extraction technologies were compared.The characterization of intracellular polysaccharopeptide was primarily identified. The resultsshowed: total sugar content was 64.41%, peptide content was 22.23%, reducing sugar content was0.22%, and its Chromatogram was consistent with that of standard. The molecular weight of itsvalid peak is 27207Da and its content is 20.60% in total, above 1.93% of standard. These indexeswere in accordance with China medicine standard WS-XG-021-2002. The polysaccharopeptide wasscanned by UV from 190nm to 400nm, the figure showed the characteristic of proteglycan. It hadα-and β-linkages by IR. At the same time, GC analysis showed that the intracellularpolysaccharopeptide was composed of arabinose, xylose, mannose, glucose, galactose. The aminoacids of the intracellular polysaccharopeptide and extracellular polysaccharide were also analyzed.The result showed that there were seventeen kinds of amino acids in them. Essential amino acids ofthem made up 29.45% and 17.85% of the total respectively.
