Coriolus Versicolor,is a rare edible and medicinal fungi which has various biological functions,however,at present,most of researches focus on the function and activation of polysaccharide,Studies on its small molecular active peptides were not comprehensively reported,therefore,this paper mainly discussed several parts around the submerged fermentation of Coriolus Versicolor and extraction of peptides with main results are as follows:1.Affect of different culture medium on yield of mycelium of Corilus Versicolor were studied.The best composition of culture medium was obtained by single factor and orthogonal design which composed of 4%sucrose,1.5%peanut meal,1.0% KH2PO4,0.5%MgSO4,0.5%Ca2SO4,0.01%inositol and its pH is 4.5,under the optimal conditions,the biomass is 1.83 g/L.2.Affect of different culture conditions on yield of mycelium of Coroilus Versicolor were studied.Through the single factor experiment and orthogonal experiment,the optimal culture conditions of submerged fermentation of Coriolus Versicolor were as follows:the temperature was 25℃,inoculation volume was 15%, shaking speed was 200rpm,the volume of medium in 250ml flask was 40%, fermentation time was 6d,under the optimal conditions,the biomass is 1.95g/L.3.Ultrahigh pressure extraction technique was first applied to extract active peptides from mycelium of Coriolus Versicolor at normal temperature,the most optimal processing parameters were determined:material-water ratiol:5g/mL, soaking2.5h at normal time,400MPa holding 2min.The yield of active peptides is 3.23%,with·OH clearance rate is 62.25%.4.The enzymolysis conditions of the three proteinases on the mycelium of Coriolus Versicolor were analyzed,then a neutral proteinase was selected,Through the single-factor experiment and orthogonal experiment,the optimal hydrolysis conditions were as follows:pH8.0,temperature 50℃,substrate concentration 10%, enzyme/substrate ration(E/S)1.0mg/g,and hydrolys 3 hours,Under this condition,the degree of hydrolysis was 33.28%.5.Activated carbon was chosed as decolouring agent of hydrolysate,it is an effective and simple decolourizer on the decolorization of hydrolysate.The result showed that high degree of discolourization could be obtained under the following conditions:activated carbon concentration 3.0%,pH4.0,temperature 50℃,time 30min,detouring and the loss rate of peptides are 86.21%and 6.28%respectively, with·OH clearance rate is 70.36%.6.The hydrolyte was filtrated by filtration,using 10kDa,5kDa and 3kDa membrane of Millipore Company,And the yield of peptides and the IC50of ofhydroxyl radical scavenging of different molecular peptides were analyzed,the results showed that MW were less than 3kDa,the yield of peptides was the highest, and the ability of radical elimination was the strongest,with the yield and IC50of active peptides are 38.67%and 1.08mg/mL respectively.
