The planting area and output of tobacco in China stand first in the world list, and thetobacco planting and production play an important role in the economic development. As agreat country in tobacco production, China yields 23 million tons tobacco a year, of which3035 percent are low-grade or unable to be made into cigarette.Solanesol is an important medium in medicine, can be used to synthesize Co-enzyme Q10and vitamin K2 . There are about 0.51.5 percent of solanesol in waste tobacco. Chlorogenicacid is also an important bio-active compound and is wildly used in food, medicine andchemical industry. It has the antibacterial, antivirus and anticancer effects. The content ofchlorogenic acid in tobacco is about 1.5%, and there have been no reports about theextraction of chlorogenic acid from tobacco yet.Taking the waste tobacco as the raw material, the determination of the solanesol in tobaccoby HPLC method, the extraction and isolation of solanesol and chlorogenic acid, and thepurification of solanesol are studied in this paper. The main achievement are as following:(1) In order to extract the solanesol from waste tobacco, the determination method ofsolanesol was established firstly. Using the HPLC instrument, column Hypersil Bos-C18 5μm4.6mmid×250mm, flow phase methanol : isopropanol=1:1(v/v), temperature 30℃,theRt(retention time) of solanesol is about 6 min and the content of solanesol in high-grade andlow-grade tobacco is 1.403% and 0.804% respectively.(2) Comparing the SFE (supercritical fluid extraction) method with the solvent extraction ofsolanesol from waste tobacco, it was concluded that the solvent extraction method is thebetter method to extract the solanesol from waste tobacco. Under the optimum conditionssolvent 95% ethanol, granularity 20 mesh,time 12 h, temperature 70℃, tobacco-to-solvent1:10(w/v), extracted two times, 93.7 percent of solanesol was obtained from tobacco.Meanwhile, the influence of saponification, and acidification were studied.(3) Through determining the chlorogenic acid in raw tobacco and its ethanol extractrespectively, it was known that the chlorogenic acid were mostly extracted out from tobaccosimultaneously when extracting solanesol under the optimum conditions. Different methodsto isolate the chlorogenic acid from ethanol extract were compared, the optimum one isplumbum acetate sedimentation method, and the recover ratio of solanesol and chlorogenicacid is 90.7% and 95.1% respectively.(4) The impurities in the solanesol crude extract were extracted by 80% ethanol, followed bycrystallizing in methanol solvent two times, the purity of solanesol was increased to 84.6%.Then the silica gel preparation column chromatography was used to purify the monomer ofsolanesol and got a white crystal. On the basis of IR, HPLC-MS spectra, the white crystalwas identificated as the solanesol monomer and its purity is 99.9%.
