High purity of solanesol is an important intermediate material of medicine. It can be applied to synthesize coenzyme Q10 Vitanmine K2 and anticancer medicine etc. however, the result of research has shown that the purity of solanesol must reach to m above 90%, according with order of synthesizing medicines. So it is nessary that new method is developped about extraction of solanesol.This dissertation studies mainly new method of extraction high purity solanesol, using the technology of supercritical carbon dioxide extraction and conbining Silica gel column chromatography. Meanwhile, mathematical model is established about he supercritical extraction of solanesol and predicted the rate of extracted solanesol are discussed. The main contributions are as follows.Supercritical fluid extraction (SFE) of solanesol was performed to study the effect of various parameters such as pressure, temperature, modifiers and amount of the optimal modifier on the yield of solanesol. an orthogonal L9(3)4test was designed, and the important factors which were selected and examined. According to variance, we further analyzed the sensitivity of the three factors on content of solanesol of the extracted. the optimal condition of the extraction was obtained, which were 25mpa of pressure, 40℃ of temperature,150ml/100g which is the ratio of 95% ethanol to tobacco.The maximum yield of solanesol was predicted to be 82.4% by established mathematical model, the predicted is consistent to the 81% yield of solanesol by experiment under the optimum condition of the extraction. Modifier enhancing technology is one of the methods which improve the efficiency of supercritical CO2 extraction. Furthemore, based on analyzing the action between modifiers and the functional ingredients, this dissertation discusses the extraction of solanesol at supercrital state which is enhanced by modifier. 95% ethanol from four modifiers is chose, as well as content and using method of modifier in the process.Because of the low purity of solanesol after the supercritical carbon dioxide extraction, it was thoroughly investigated that the condition of silica gel column chromatography for high purity of solanesol to meet the demand of systhesis. The appropriate mobile phase is decided, the mix with acetidin and petroleum ether (60/90) which proportion is 1:4.and appropriate velocity of mobile phase is 2ml/min. the rate of height and dimeter is 15:1. After separation of silica gel column chromatography,The purity of solanesol is improved from 30% to more than 94%. And recovery of solanesol is 78%.Therefore, new method is proposed to extract high purity of solanesol from discard tabbaco. The purity of solanesol is analyzing by high performance chromatography, which is applied to synthesize medicines. Meanwhile structure of solanesol molecule is analyzed by FTIR.
