Taxol is a kind of terpene compounds, which was separated from Taxus brevifolia originally. Because of the special anticancer mechanics and special curative effect for some kinds of tumor, Taxol attracts much attention recently. It is very difficult to separate and purify Taxol from Taxus, because the content of the Taxol in Taxus is very low, and there are also some analogues of Taxol in Taxus, which make it more difficult to separate. Consequently, it is very important to develop a method to separate and purify Taxol with high recoveries.Three main parts were divided in this treatise. In the first part, the method of separation and purification for Taxol and Cephalomannine from Taxus cuspidata was studied. It included one normal-phase liquid chromatographic process (NPLC) and two reversed-phase liquid chromatographic processes (RPLC). In the RPLC process, a novel high molecular resin (PRP-6) was applied. In the second part, in order todetermine the content of Taxol , Cephalomannine and 7-epi-taxol in Taxus cuspidata, a method of solid phase extraction-high performance liquid chromatography (SPE-HPLC) was developed in which the PRP-6 was used as the solid phase of SPE. The method was fast, simple and with high recoveries and repetition. In the last part, the behaviour of Taxol and Cephalomannine in thin layer chromatography (TLC) was studied, in order to inspect elutions of NPLC and RPLC. At last, two TLC methods were used to determine Taxol and Cephalomannine in elutions. The result was satisfactory.In the process of separation and purification for Taxol and Cephalomannine the NPLC and RPLC were both used. The plant extract was concentrated, then extracted by dichloromethane-water. Subsequently preparative liquid chromatographic columns were used to separate and purify Taxol and Cephalomannine in the organic phase. After through a NPLC column and twice RPLC column, two kinds of white acicular crystals were obtained, and proved to be Taxol and Cephalomannine by HPLC, 'H-NMR and 13C-NMR, and their purity were above 98%. The yields of Taxol and Cephalomannine were 0.0022 % and 0.0018 % of dry leaves of Taxus Cuspidata and the recoveries were above 70%.
