A system of fluorescence resonance energy transfer composed of Au-DNA and CdTe-DNA was built and researched.Two sizes of Au nanoparticles were prepared by reducing gold chloride with sodium citrate. The transmission electron microscopy, UV-Vis spectroscopy and fluorescence spectroscopy were carried out.CdTe semiconductor nanoparticles were synthesized in water. The transmission electron microscopy, UV-Vis spectroscopy and fluorescence spectroscopy were carried out.The overlap between emission spectrum of the CdTe nanoparticle and the absorption of Au nanoparticle was confirmed by the spectroscopy research of CdTe and Au nanoparticles. Au nanoparticles were linked to 3'-SH-DNA through Au-S. The effect of electrolyte was discussed. Then the UV-Vis spectroscopy was carried out. CdTe nanoparticles were linked to 5'-NH2-DNA through -COONH-. The effect of reaction buffer and EDC were discussed. Then agarose-gel electrophoresis was carried out. The fluorescence of CdTe-DNA CdTe-DNA-Au and CdTe-DNA-Au+target DNA were measured . Data showed that the fluorescence of CdTe-DNA-Au conjugates is minimum. When we added a complementary ssDNA to CdTe-DNA-Au conjugates, the fluorescence of the hybrid molecule increased. So the system of fluorescence resonance energy transfer was set up successfully.
