High purity phosphatidylcholine (PC) not only can prevent the occurrence of hepatitis, fatty liver and diseases in heart and blood vessel, but also has functions of lowering cholesterol amount and anticancer. In this text, we studied the separation and purification technology of high purity PC from soybean degummed oil.Silica column chromatography was selected as a primary method of the purification of soybean PC from soybean degummed oil. At this step, mixed solvent of petroleum ether (60~90℃), 2-propanol and water (1/1/0.175,V/V/V) was the mobile phase selected. Thus, if flow rate was 1.1 BV/h, soybean degummed oil/silica (w/w) was 0.5, the product purity was over 80%, and the recovery was over 90%.High purity PC was obtained by alumina column chromatography. We selected the chloroform-methanol (9/1 and 1/9) as mobile phase. When flow rate was 1.5 BV/h, raw product made by silica column chromatography/alumina was 0.06 (w/w), the product purity was over 95%, and the recovery was over 95%.To reduce the water content of soybean PC product, the vacuum freeze-drying of soybean PC was conducted. The effects of pre-freezing temperature, water content of raw material, drying time and loading capacity on the water content of freeze dried PC were studied. The optimum operation conditions were preliminary freeze temperature -35℃, the water content of raw material 50%, drying time 7 h and loading capacity 8.1 kg/m~2. The water content of freeze-dried product was less than the water content of the commercial product made by German Lipoid GMBH.
