In this thesis, Organic Assistant Peptide(OAP) were synthesized at first time by Fmoc-solid-phase method. The performance under various experimental conditions as connection rate, Deblock and Cleavage was studied. The synthesized peptide was separated and purified by semi-preparating RP-HPLC. Identification was performed by MADI-TOF mass. The method of synthesis and purification of OAP was established finally. Under the optimized conditions, OAP of purity 99% can be obtained in the yield of 36.5% compared to theoretical value.The thesis includes the following four parts:1. At first, the fundamental knowledge of peptide synthesis technology was introduced. Then the principle of peptide analysis and purification was referred. At last the application and research prospect of Bioactive peptide was also briefly reviewed.2. The OAP was synthesized by Fmoc solid-phase method. Wang resin and 2-Chlorotrityl chloride resin were both used in synthesis, the synthesis results was compared. Through the exploration of the performance under various experimental conditions, the best synthesis result was achieved. The molecular weight of synthesized peptides was identified to be correct by MALDI - TOF mass analysis.3. The separation and purification of OAP synthesized by two resin were carried out with reversed-phase liquid chromatography. The chromatographic conditions were optimized. The semi-preparation by RP-HPLC was tried. The purified products were analyzed by RP-HPLC and MALDI-TOF mass.4. The experiment results and sense were summarized. Following working schedule was listed.
