With the increasing improvement of living standard, people become to pursuit the medicine , functional food nature and health. Many researches indicate that flavones in natural product have the pharmacologicals of blood pressure depressing, lowering blood lipid, and inhibiting platelet aggregation. So it was taken into account by governments, pharmaceutical and food industries. The extraction of flavones from natural product was the key point of its research and development. In the chapter I, authors summarized several kinds of analysis and extraction technologies in recent years and discussed its review of application.In the chapter II, micellar electrokinetic capillary chromatography (MECC) was applied to the simultaneous separation and determination of six derivants of 3, 4-dihydropyrimidin-2-ones first time. The optimal conditions of electrophoresis such as the pH and concentration of buffer solutions is studied. The six analytes could be well separated by using 5 mmol/L sodium borate - 75 mmol/L SDS-20% (v/v) methanol + isopropylalcohol (Vmethanol: V isopropylalicohol = 1:4), pressure injection (0.5 psi) time 5 s, voltage 20 KV and temperature 25 ℃.This method is a rapid, simple and useful technique for identification, separated and determination of six derivants of 3, 4-dihydropyrimidin-2-ones.In the chapter III, the simultaneous separation and determination of eight derivants of 3, 4-dihydropyrimidin-2-ones were investigated by different electrophoretic methods. The sample is determined by partial-filling micellar electrokinetic capillary chromatography (P-F MECC) and conventional micellar electrokinetic capillary chromatography (MECC) and found the proper electrophoretic separation conditions. Electrophoretic separation conditions are optimized. Six components are separated completely. The peaks of others are partial overlapped. P-F MECC is favorable method for biomedical analysis because of its high efficiencies, short analysis time and minimal sample and solvent requirements.In the chapter IV, extraction process of flavonoids from buckwheat pollen byultrasonic wave and refluxing with methanol were tested. The rutin was used as a standard compound, and the optimum extracting conditions of flavonoids were investigated by using orthogonal experimental design. The optimal conditions for ultrasonic wave were 40 KHz ultrasonic wave, 15:1 of methanol to pollen weight, extraction for 140 min;and for refluxing with methanol were material ratio 15:1, extract for 70 min at 80 °C. Compared with ultrasonic wave extraction, refluxing extraction was more effective with advantages of short extracting period, higher yield and simple equipment. So it was a useful method for extracting flavonoids from buckwheat pollen. In the chapter V? the content of flavonoids in buckwheat pollen was investigated by HPLC. Flavonoids were extracted by refluxing with methanol. Quercetin, isorhamnetin in extracts was determined by HPLC after the hydrolysis of all flavonol glycosides into aglycones. The used column was Shima.Packvp-ODSCi8,150 mm X 4.6 mm(5 um );The mobile phase consisted of solvent A (methanol) and solvent B (phosphoric acid, pH=5.12) for gradient elution. The flow rate was 1 mL/min and detection was effected at 368 nm. The results show that the content of quercetin and isorhamnetin in every gram of buckwheat were 0.49 mg, 0.063 mg, respectively.
