By separating on deferent samples in this experiment,we got one more efficient strain on cellulose's decomposition,via primary identify we thought it was Trichoderma viride .Subsequently,we studied on its solid state fermention,and optimized its conditions of cellulase's production,the results as underside: the proportion of rice straw powder to bran was 3:2,the proportion of water to material in medium was 2.5:1,the optimum nitrogen sourse was 0.5% (NH4)2SO2, the optimum cultivate time was three days,temperature 30℃, 10% inoculating volume,initializing medium PH 6.0-7.0.Under these conditions,the activity of cellulase of dry substrate per gram can achieved 18.4FPA U/g and 56.7 CMC U/gStudies on the cellulase which the Trichoderma viride yield indicated: the cellulase activity arrived its tiptop to filter paper when PH was 5.2,and pH4.8 to absorbent cotton.The optimum hydrolyze temperature was 55℃ to filter paper and 50℃ to absorbent cotton.At 80℃, cellulase almost has no activity, and after pH8, the cellulase activity reduced rapidly. We also found that ethanol can restrain the cellulase hydrolyze,so we should release from it in actual production.It has a tight relation on H2O and intensity of cellulase's hydrolysis in the course of decomposition.The stronger of hydrolysis,the more of H2O in medium,it reached its tip top on the fifth day,later,it became little and little.Every kind of enzyme's cellulase activity are different in the hydrolysis course.Activity of endo-β-1,4-glucanase upswung quickly,it reached its tip top on the third day,subsequently descend day after day.
