Study On Preparation Of Geqin Microcapasual

Geqin microcapsule is a new compound Chinese medicine, which consists of Radix Puerariae and Radix Scutellariae. Pharmacodynamoics studies indicate that it can strengthen protective effects of ischemic preconditioning on myocardium, reduce the myocardial infarct size and incidence of ventricular arrhythmia. It has favorable results on the prevention and therapy of cardiovascular diseases. The dosage form of drug was chosen as microcapsule for the demand of clinic, the function and treatment as well as the characters of component of the herbs. In this paper we studied the preparation of Geqin microcapsule.The optimum extract conditions were selected by applying orthogonal test design and grading methods of synthesizing multiple guidelines. The suitable condition for technology of Geqin microcapsule is extracting with 70%EtOH twice ( 2 hours every time, 10 times for the first and 8 times for the second), purifying with clearing medium and AB-8 macro porous resins which make the content of total flavonoids reaching 57.40%. The content of total effective components in the compound reaches 50%, which meets the requirement of new drug applications. Geqin microcapsule was prepared by simple coacervation method. Using the uniform design, the preparation technique of Geqin microcapsule was optimized, and the quality was evaluated. The optimum preparation technique of Geqin microcapsule: the ratio of core material to coating material was 1:2, the temperature of encapsulation was at 55°C and the stirring speed was 1200r/min. The results showed that the preparation technique of Geqin microcapsule had well entrapment rate, appropriate diameter, distribution of microcapsule particles size and dissolution test ,which met the requirements of Appendix Guiding Principles of Microcapsule, Microsphere and Liposome in Chinese Pharmacopoeia(2000 Edition).Now ischemic preconditioning is the most effective in protectingmyocardium by itself. Thus the result made a good base for Geqin microcapsule that would be the new drug of strengthening protective effects of ischemic preconditioning on myocardium.
