This article selected containing isoflavone and flavonoids commonly used in traditional Chinese medicine radix puerariae,radix scutellariae, to investigate the effect of chitosan on the purification of isoflavones and flavonoids.Settling the single herb radix puerarin , radix scutellariae on the water extraction liquid with chitosan , L9(34) orthogonal test was first used to determine the best water extraction process of the puerarin and baicalin; then chose the best clarify technology of radix puerarin, radix scutellariae's water extraction liquid using chitosan flocculating sedimentation method by the L9(34) orthogonal test method. The results showed that dosage of chitosan had significant influence on the effective components of radix puerarin and radix scutellariae among the factors for concentration, dosage of chitosan and the pH value. Through comparing the settlement of effective ingredients of content, yield of extract and the original solution fingerprint similarity, determine the chitosan flocculation settlement method to the clarifications process of radix puerarin and radix scutellariae's water extraction liquid is steady and feasible. In addition, purified radix puerarin and radix scutellariae medicinal material water extraction fluid by alcohol sedimentation, natural settlement method and high speed centrifugation, With puerarin and baicalin respectively for index comparison of purification effect of different purification methods, results to determine chitosan flocculating sedimentation method to the water extraction of Radix puerariae and Radix scutellariae is superior to other three methods of liquid, stable and feasible. Herbal prescription of gegenqinlian decoction contain Radix puerarin, Radix scutellariae and rhizoma coptidis, According to the single herb for trial operation, gegenqinlian fluids carry liquid with chitosan flocculating sedimentation and alcohol precipitation. Comparison of compound effective ingredient content, the size of extract yield and the original solution fingerprint similarity, it is concluded that the clarification process of the settlement of gegenqinlian decoction compound method also reliable and practical.
