SCFE - CO2 technology was studied systematically and deeply, using mulberry seed oil as raw materials in this paper. On the basis of single factor experiments, mathematical model between the technology parameters and extraction efficiency was founded by quadratic general rotational combination design experiment of four factors, the operative parameters was optimized by calculation and analysis of model. The optimum extraction technology was chosen by comparison of SCFE - CO2and ultrasonic extraction, microwave extraction,Soxhlet extraction, impregnant marinating. Physical and chemic quality , fatty acid, V E and microelement etc of mulberry seed oil was analyzed systematically to open out the relation between the operative parameters and quality of mulberry seed oil. Dynamic transfer process model was founded by scanning electron microscope analysis of extracted remainder of SCFE - CO2mulberry seed oil.The result as follows:1. Components of Mulberry seed: water 4.60%, crude lipin 31.48%, protein 28.72%, ash 2.39%, crude fibre 28.37%; Physical and chemic quality of mulberry seed oil, water 0.75%, density 0.9210, colour and luster Y25,R0.2, clarity, refraction exponential(n20)1.4744, acid value 0.75mg/g,peroxide value 4.06meq/kg,iodine value 130.35 g/100g.2.The optimal parameters getting by single factor experiment are, water content of mulberry seed 6%, loading 250g, granule diameter 0.9mm, extraction pressure 30MPa, extraction temperature 40℃, extraction time 110min, CO 2flowrate 10L/h, pressure of separatingⅠ7.5 MPa, temperature of separatingⅠ35℃, pressure of separatingⅡ4~6MPa, temperature of separatingⅡ28℃.On the basis of relationship of pressure of extraction and temperature of extraction , the conversion pressure of system was ascertained ,result of which is 27MPa.Under the conversion pressure, extraction ratio declined with increasing of temperature of extraction, on the contrary increased.3.The optimized the operative parameters of quadratic general rotational combination design experiment of four factors: pressure of extraction 37MPa, temperature of extraction 50℃,time of extraction 130min, CO2 flowrate 6L/h, mathematic model is correct by analyse. Discussing of regression coefficient indicated that all coefficients have evidently effect besides b3 4, b1 1. Getting the result that every factors have different influence to ratio of extraction by main effect analysis, x2 > x4>x3>x1, interaction of all factors is notable besides x 3x4.
