Adenine, which exists in the nucleic acid of the organism as the base, has drawn great attention of biochemists and organic synthetical chemists in recent years because of its good active biologic quality. Especially in the respects of antivirus and restraint of cancer cells, adenine derivatives have obvious effect and alluring prospect of clinical application. This thesis starts the synthesis and research of compounds with active biologic quality by using adenine as the basic material.First of all, the analogic compound of anti-hepatitis medicines—adefovir, that is N6-benzoyl-9-[2-(phosphono methoxy)ethyl]adenine is synthesized. On the basis of previous researches, route of reaction is designed and confirmed, and the Adefovir analog is synthesized through 11 steps containing parallel reactions that include esterification, contraction, protecting group, substitution, and hydrolysis etc. by using glycol and adenine as the materials. All reactions are classical. The synthesis of adefovir is hopeful to be industrialized because of its mild conditions, easy acquiring of materials and reagents, and a little bit high productive rate. The conditions of reaction and purification methods of this route have been improved and optimized greatly compared with the previous work.Besides, we also use N6-benzoyl-9-(hydroxyethyl) adenine...
