Objectives To establish a method for the determination of perfluorooctanoic acids (PFOA) in water and urine. To investigate the adsorption and the photochemical degradation of PFOA in aquatic environment. Methods PFOA was extracted as an ion pair with tetrabutylammonium (TBA) ion into methyl tetrabutyl ether(MTBE) and the ion pair was derivatized with ω-bromoacetophenone(ω-BaP). Then the analyte was determined by HPLC with UV detector and external standard method. The contents of PFOA in adsorption and photochemical degradation study were determined by proposed method and the adsorption rate and degradation rate were calculated respectively.Results: The determination conditions were optimized as follows: The chromatographic conditions were Supelco C18 column (5um, 250×4.6mm); column temperature was 25℃; mobile phase was methanol-water; flow rate 1.0ml/min; gradient elution from 0 min at 80% methanol-water into 100% methanol within 8.0min, then go on for 5.0min to elute other impurities. The extraction conditions were: 0.25mol/L TBA 1.5ml, 4.0ml MTBE twice; The derivatization conditions were: 10mg/ml, 25 mg/ml, 60mg/ml ω- BaP 10μl derivatization temperature, 80℃; derivatization time, 80min.
