Lycopene as one of the carotenoids is an efficient antioxidant. Extraction of lycopene from Blakeslea trispora biomass was presented in the paper. It contains the pretreatment,extraction and purification scheme of lycopene. According to these studies , we promoted the industrial production scheme of lycopene.1)A new method of thin layer chromatographic (TLC) determination of lycopene was developed. The optimal conditions of TLC were as follows: silica gel G as stationary phase, n-hexane-dichloromethane (9:1, v/v) as mobile phase. The Rf value of lycopene andβ-carotene was 0.45 and 0.63.2)A new method of reverse phase high performance liquid chromatography (HPLC) determination of lycopene was developed. The HPLC conditions were: Alltima C18(4.6 mm×250 mm, 5μm)column as stationary phase ,methanol-THF-water(70 : 26 : 4,v/v/v) as mobile phase ,the flow rate was 1.5 mL·min-1 ,column temperature was 30℃, UV-vis detector (472nm). The calibration equation was Y = 1.32×108 X– 1.81×105, the liner range for the content of lycopene was between 0.072 and 0.36μg , r = 0.9999, the recovery of lycopene was 98.61% to 100.46% and the RSD was 2.23%( n = 5). When the signal-to-noise ratio was 3:1, the limit of determination was 7.34×10-7μg.3)The pretreatment of lycopene extraction was studied in the paper. The wet biomass were deal with 95% food grade alcohol to remove water. The optional ratio of wet biomass to alcohol was 1:1(g/ml). The de-water biomass were dried at 45℃for 2.5 hours. The extraction was consequently done after the dried biomass were treated by miller for 30minutes with acetone.4)The extraction of lycopene was developed. The effects of extraction temperature, extraction time, extraction times and liquid-solid ratio on the lycopene extraction were developed. The results showed that extraction temperature, extraction time and liquid-solid ratio were major factors. We developed the effect of n-hexane, ethyl acetate and acetone on lycopene extraction, and the optional extraction conditions were: n-hexane as solvent, extraction temperature 40℃,extraction time 1hour and liquid-solid ratio 1:40. Based on the above extraction conditions, more than 80% lycopene could be obtained.5)The purification process of lycopene was developed.
