Extraction Of Diosgenin From Dioscorea Zingiberensis

As the main active ingredient of D.zingiberensis Wright, diosgenin is the important raw material for the synthesis of steroid hormone medicine and steroid contraceptive medicine. In order to improve the yield of diosgenin, the abstraction of diosgenin in D.zingiberensis Wright was studied by five methods, such as traditional hydrochloric acid, fermented, phosphoric acid, two-step extraction and microwave abstraction. The influence of preparation techniques on abstraction yield was mainly discussed in this paper. And the products were analyzed by a series of means, such as melt point, element analysis, infrared, nuclear magnetic resonance, etc. In addition, the waste water produced during the abstraction process was simply treated. The following conclusions were obtained:1) Five methods such as traditional hydrochloric acid, fermented, phosphoric acid, two-step extraction and microwave abstraction were used to abstract diosgenin in D.zingiberensis Wright. The experimental results show that the yield of two-step extraction is highest, and the quality of diosgenin is also relatively better. The yield of diosgenin drawn by this method is up to 2.41% and has improved by 9.55% than the method of hydrolyzing only directly(2.20%).2) In this paper, the method of two-step acid hydrolysis on extracting diosgenin from D.zingberensis wright was chiefly researched by orthogonal test. The results show that relatively better experimental condition is 20 ml,1 mol/L HCl,acid hydrolysis for 3h.3) The application of microwave technology gets very great development in recent years. The traditional hydrolysis and microwave extraction were carried on to extract diosgenin from the D.zingiberensis Wright. The yield of diosgenin extracted under microwave for 20 min is 1.92%,which is little lower than soxhlet extraction for 5 h.
