Fructose and fructose-6-phosphate is the important material in organism metabolism process. To accurately quantitative analyse above mentioned material in different medium such as in buffer aqua, water and real life is one of the key techniques in analysing interim product in Metabonomics and Metabolism Engineering.Fructose or fructose-6-phosphate is changed to α -hydroxymethylfurfural under the hydrochloric acid(HCl) catalyst condition. Then produce UV-Vis spectral absorbance. While add Benzodiazepine to the above-mentioned reaction, it will produce a red anthracene of xanthone derivatives, which enhancing the UV-Vis spectral absorbance. So we mainly choose above two conditions in this research to do quantitative analysis by using UV-Vis spectrum, and make a quantitative analysis of fructose, fructose-6-phosphate and its related material such as glucose, glucose-6-phosphate in buffer aqua or water, the purpose of using buffer aqua is to simulating the complicated composition of life or environment.In the quantitative analysis of UV-Vis spectrum, we analyse the linear range of standard aqua on one side, and adopt whole wave band, absorption peak and the wave band of a high correlation of the absorbency and concentration to do Multiple Linear Regression Analysis on the other hand. Cross-validation mode was used to do quantitative analysis of fructose, fructose-6-phosphate, glucose, glucose-6- phosphate and the binary systems of fructose and glucose, fructose-6- phosphate and glucose-6-phosphate by using UV-Vis spectrum. Did four kinds of total 24 groups of cross-over trial under four condition with Tris-HCl/Resorcinol, Tris-HCl, Resorcinol and water four conditions. Various quantitative analysis modes were used to analyse each experimental result in order to obtain the optimization quantitative analysis method. Finally, above-mentioned method were applied to do a quantitative analysis of sugar cane juice, and also do quantitative analysis of Mid-Infrared Spectroscope (FT-IR/ATR) in contrast.
