Fructose is one of the most common ketose.Found in honey and fruit,sucrose and glucose combined with daily consumption.Because not easy to lead to high blood sugar,and not easily get fat and fat,more won't produce cavities,which is used by more people.At present,the developed countries have fructose is widely used in food industry,medicine and health products production.And fructose in China is not widely used and actual production capacity is insufficient,more fructose in F42 type is given priority to,main sources of fructose in China depend on import.This experiment by broken rice as raw material,by means of single factor experiments and multi-factors orthogonal test analysis,SAS,data analysis and other methods,enzymatic liquefaction is studied in this paper,the preparation of glucose enzymatic saccharification process parameter optimization,fructose syrup isomerization preparation technology parameters optimization,simulated moving bed separation of high purity crystalline fructose fructose process conditions optimization and solvent method process conditions.Don't use chromatography,HPLC method such as identification of fructose.1.The broken rice as raw material preparation of glucose,the optimal process of research.This experiment through alkali extraction with broken rice starch row finishing,and the DE value of glucose to experimental indexes,through the check material liquid ratio,alpha amylase enzyme amount,reaction time,liquefaction temperature,p H value environment five factors influence on liquefaction effect,optimum parameters of each single factor determining liquefaction is: the material liquid is 1:6,alpha amylase enzyme amount for 15 u/g,the reaction time of 25 min,liquefaction temperature is 90 ?,the natural pH value,under the condition of the measured the DE value of liquefaction liquid was 18.6%.Glucose of amylase were studied with enzyme amount,saccharifying environmental pH value,reaction time and the saccharification temperature the effects of the four factors on the saccharification liquid to determine the optimum parameters of saccharification factors as amylase and glucose enzyme amount is 120 u/g,saccharification environmental pH value is 3.5,the reaction time is 24 h,saccharification temperature was 60 ?,saccharification liquid can be measured under the reaction conditions of DE value of 99.3%.Liquid sugar use diatomite decoloring again after two consecutive after ion exchange resin series of Yin and Yang,pH 7.0,the color value is 0(IU).After saccharification liquid refined colorless,and almost no impact on the quality of syrup.2.Enzymatic isomerization preparation F42 research of fructose syrup.This experiment by enzymatic isomerization of producing fructose syrup process,fructose content as experimental indexes,studied the glucose isomerase plus enzyme amount,pH heterogeneous environment,isomerization temperature,reaction time and so on four factors on the influence of fructose content,determine the scope of the isomerization of each single factor changes,on the basis of single factor experiment data,the orthogonal experiment analysis and SAS statistical analysis of isomerization from fructose syrup process by line optimization,the process parameters were optimized as follows: glucose isomerase with enzyme amount4mg/g,pH value of heterogeneous environment is 8,and isomerization temperature is 60?,the reaction time is 48 h measured under the reaction conditions of fructose syrup fructose content was 43.7%.Fructose syrup after refining,the measured pH value of 7.0,the color value is 0(IU).Fructose syrup after refined colorless,don't and there isalmost no impact on the quality of fructose syrup.3.The simulated moving bed separation research of high purity fructose.This test use the safety factor method is applied to the simulated moving bed,to achieve the separation of fructose and glucose.(1)by single pulse test,separating degree as index,the single factor experiment was carried out,get a drift Wright SPC-106ca2 + resin has better adsorption effect,the best injection conditions: column temperature was 60 ?,the concentration of the sample was 60%,the elution flow rate of 1.5 mL/min,sample quantity of 15 mL.Diction of using single pulse test results,the SMB is obtained by safety factor method of simple parameter design method,the theoretical parameters of simulated moving bed.Of simulated moving bed(3)through the actual operation,to the theoretical value is optimized in the experiment,get the optimal preparation of high purity fructose SMB parameters: the switch time of 480 s,feeding velocity of 0.37 L/h,eluent,flow rate of 1.27 L/h,extract velocity of 1.03 L / 0.82 L/h h,raffinate velocity,circulating flow rate of 1.98 L/h.Under the condition of the operation,fructose 99.54% purity,yield was 83.95%,the purity of glucose was 90.73%,the yield of89.38%.4.The crystalline fructose process research.This test using the method of solvent crystalline fructose,first through the single factor experiment,determine the scope of the various factors.On the basis of the orthogonal combination experiment and SAS statistical analysis of the crystallization process of fructose condition in optimization.Get the best process parameters for syrup concentration ratio of 1:10 and anhydrous ethanol 80%syrup for 65-5 ?,the temperature difference,the crystallization time of 72 h.Measured under the conditions of fructose 99.95% purity,yield 87.94%.
