Lentinus edodes was used as research material and the water-soluble polysaccharides extracted were explored in the following aspects:1. An enzymatic method for analysis ofβ-glucans, which are effective constituents in polysaccharides from Lentinus edodes, has been studied. Thus, the quality control could be carried on to the raw material. After hydrolyzing by thermo stableα-amylase, papain and amyloglucosidase, the content of glucose was measured by GOD-PAP method and that ofβ-glucans was computed. The dosages of the three enzymes were determined and the content ofβ-glucans in the samples were in the range of 3.70%~3.85%. The average recovery was 98.7%. The RSD of precision is 1.61%(n=9).Water-soluble polysaccharides were extracted from fruit body of Lentinus edodes with hot water. The extracts were precipitated with ethanol, and purified by Sevag method and macroporous resin. The ratio of Sevag reagent to material is 5:2, and the reagent was used 7 times to remove protein. H2O2 and macroporous resin were compared in the effect of discoloring. AB-8 macroporous resin was chose in the purification process. The purified polysaccharides were further isolated by Toyopearl HW-75 gel filtration column chromatography and two fractions were obtained successfully, named as XG1 and XG2. The yields of the two fractions are 0.83%, 1.20%, respectively.2. The basic structure analysis of XG1 and XG2 were studied using several chromatographic and chemical methods. UV and high performance gel permeation chromatography (HPGPC) were used in identifying the chemical homogeneities of .XG1 and XG2, and the molecular weight of which were determined by HPGPC. Determination conditions were as follows: column: Shodex Ohpak SB-805 HQ, Shodex Ohpak SB-804 HQ and Shodex Ohpak SB-803 HQ in series. mobile phase: NaH2PO4-Na2HPO4 buffer; flow rate: 0.4mL/min–1; detector: refractive index detector (RID).
