The polysaccharides BJPA, BJPB1, BJPB2 have been isolated and purified from coarse-polysaccharides(A and B)by using distilled-water as eluant in Sephadex G-150 column chromatography. coarse-polysaccharides(A and B) were obtained from BAO JI PILL by water-extracting, alcohol-precipitating. Single peak shape was showed in Sephadex G-200 column chromatography. Their molecular weight were respectively estimated to be 5.2 X 104, 9.2 X 103, 3.5 X 103.BJPA, BJPB1, BJPB2 are water-soluble and heterogeneous polysaccharides. The composition of monosaccharide have been identified by acid hydrolysis and HPLC analysis. The structure have been analyzed by IR, periodate oxidation, HPLC and Smith degeneration. The results indicate that BJPA consists of L-Rhamnose (38.3%), D-(+)-Xylose (15.3%), D-Mannose(18.2%), D-Glucose (12%), D-Galactose (16.2%); BJPB-1 includes L-Rhamnose (4.0%), D-(+)-Xylose (6.7%), D-Fructose (19.0%), D-Glucose (72.6%); BJPB2 includes L-Rhamnose (15.4%), D-(+)-Xylose (22.5%), D-Glucose (48.1%) and D-Galactose (14.0%). The IR spectrum indicated that BJPA, BJPB1, BJPB2 are linked by a - glycosidic bonds, while the backbone of BJPA is mainly linked by 1â†'2 glycosidic bonds, with a few 1 â†'6 linkage, BJPB1 and BJPB2 are mainly linked by lâ†'6 glycosidic bonds, with a few 1 â†'2 linkage, according to the results of periodate oxidation and Smith degradation.
