Lactone compounds, with the same nuclear structure as that of phthalide, are the bioactive components of Ligusticum Chuanxiong Hort. Recent researches indicate that lactone components in L. Chuanxiong Hort can improve the cerebral blood flow through inhibiting thrombus formation and platelet aggregation during ischemic attack, and have exhibitted promising effects as preventive and therapeutic agents for antiatherosclerosis.The chief research purpose of this dissertation was to develop the extraction and isolation technics for the process preparation of lactone compounds from L. Chuanxiong Hort. The total lactone compounds in the final products should be higher than 50%, so as to suffice the requirement of SFDA for the new drug application. The main research results showed as follows:1. To have developed the method to simultaneously determine senkyunolide-H, senkyunolide-I, sedanenolide and Z-ligustilide, four main lactone compounds in L. Chuanxiong Hort, by reversed phase high performance liquid chromatography ( RP-HPLC ), and also the method to determine total contents of lactone components in Rhizoma Chuanxiong by UV-spectrophotometry.2. The factors affecting the extraction rate of lactone components from L. Chuanxiong Hort, appropriate extraction conditions have been obtained from the single-factor-experiment and orthogonal array design (OAD) experiments. Rhizoma Chuanxiong was hydrolyzed by three enzymes. Such treatments may enhance senkyunolide extraction at the industrial level as well. Lactone compounds in Rhizoma Chuanxiong were extracted with mixed solvents.The operation is simple and the cost is low. All of the four components from the lactone extract were stable when being stored at low temperature.3. To have combined the optimized extraction and isolation conditions in lab with Pilot-scale experiment, the best technical preparation conditions of total lactones from Rhizoma Chuanxiong have been confirmed.
