Ligusticum Chuanxiong Hort. is one of Chinese traditional drugs with the function of promoting flow of Qi and Blood and removing wind for stopping pain. As a drug, it can be traced back to Shen Nong Ben Cao Jing. Ligusticum Chuanxiong Hort. is distributed extensively in our country, especially in Sichuan province, and its resource is abundant. In this paper, polysaccharide in Ligusticum Chuanxiong Hort. is studied, about which there was no reports as yet.There are two parts in this thesis:Part one: Review1. The evolvement of chemical component and pharmacological activities of Ligusticum Chuanxiong Hort. More than 263 kinds of chemical components have been found in Ligusticum Chuanxiong Hort., including essential oils, lactones, phenols, organic acids, inorganic elements and so on. The studies on the pharmacological activities of Ligusticum Chuanxiong Hort. are widely involved in the cardiovascular system, digestive system, urinary system, respiratory system, nervous system, reproductive system and immune system.2. There are three faces in the research advancements of polysaccharides: the history of the studies on polysaccharides, a review on the situation of the pharmacological activities of polysaccharides, the application and the development direction of the studies on polysaccharides.Part two: Research papersIn this part, five departments are involved.First, extraction, isolation and purification of the polysaccharides in Ligusticum Chuanxiong Hort.. The sample was defatted with petroleum ether, removed the essential oils with steam distillation. Then the water-soluble polysaccharide was etracted by boiling water from the sediment, and then was precipitated by 80%(v/v) of ethanol. The crude polysaccharide in Ligusticum Chuanxiong Hort. obtained is maroon. Using the method of Sevag, the protein in the product is removed. Then compounds with small molecular weight were removed with dialyser. In the end, the product was precipitated by 80%(v/v) of ethanol again. The precipitated was washed by 95%(v/v) of ethanol, ethanol, acetone and aether in order. Thus we obtain a part-pure product of polysaccharide in Ligusticum Chuanxiong Hort.(LCP). LCP is separated into fourfractions by DEAE-cellulose chromatography, which was gradient eluted by water, 0.1, 0.2, 0.3 and 0.4 mol-L-1 of NaCl in order. And LCP-1, LCP-2, LCP-3 and LCP-4 are obtained, respectively, in water, 0.2, 0.3 and 0.4 molL-1 of NaCl solutions. The yield rates of them are 35%, 3.6%, 18% and 26%. Then they are purified by Sephadex G-200 gel chromatography and tested by gel filtration on Sephadex G-200 and UV spectrum. Typical absorption of nucleic acids or protein cannot be found in the UV spectrum and there is a single symmetric peak by gel filtration, respectively, which implies they are homogeneous component.Second, a preliminary analysis of the structure of the polysaccharides in Ligusticum Chuanxiong Hon.. Their average molecular weight of the polysaccharides is approximately 31 kD, 52 kD, 90 kD and 36 kD in order, determined by gel filtration. LCP-1, LCP-2, LCP-3 and LCP-4 are hydrolyzed by 2.0 mol.L-1 of sulphuric acid, and then their components and structure are measured by GC chromatography. The results show that LCP-1 is mainly composed of glucose, LCP-2 is consisted of arabinose, galactose, mannitose and glucose, the same as LCP-3, and the main composition of LCP-4 is arabinose. LCP-1, LCP-2, LCP-3 and LCP-4 have typical characteristic absorption peaks of polysaccharides in their 1R and 'HNMR spectrum, which indicates alpha-glucosidal bond in LCP-1, alpha-glucosidal and beta-glucosidal in LCP-2, LCP-3 and LCP-4. The same result was obtained in the 13CNMR spectrum of LCP-1.Third, flow injection on-line hydrolysis spectrophotometry for the rapid determination of polysaccharide in Ligusticum Chuanxiong Hort.. The determinations of polysaccharides are commonly realized by the determination of reduce sugar, which was hydrolyzed from polysaccharides in advance. Those methods are very tiring, time wast...
