Immobilized Metal Affinity Capture Protein Chip is an extra-ordinary valuable tool for separating and purifying protein. Combining Surface-Enhanced Laser Desorption and Ionization Mass Spectrometry (SELDI), it is one of the currently used techniques to identify biomarkers for cancers. Studying and preparation of carriers for protein chip has an important actual meaning to popularize the use and enhance the performance of protein chip.RF magnetron sputtering, sol-gel spin-coating and thermal oxidation were used to prepare SiO_x film on Al substrate and silicon wafer. The surface morphology, structure and bonding properties of SiO_x film were studied. The results show that the film deposited by R.F. magnetron sputtering on Al substrate etched in NaOH solution had rough surface morphology and the film is bonded well to the substrate; The film obtained by sol-gel spin-coating on smooth Al substrate had porous surface morphology without crack after using DMF and the film are also tightly combined. The film obtained on smooth, porous, textured silicon wafer etched in chemical solution by thermal oxidation had different surface morphology and very tight combining capability.Immobilized metal ion affinity methods were used on SiO_x film with smooth, porous, textured surface morphology obtained by thermal oxidation on silicon wafer to modify the surface. Several influencing factors to protein adsorbing were studied. The results show that the textures had well adsorption ability with protein. The process of aldehydation during surface modification was important for keeping more polar atom which can absorb protein. Different kinds of metal ion chelater with dissimilar PH value had distinct adsorbing ability.
