Synthesis of hydantoin derivatives and application of triphosgene incarbonylation reaction have been studied in this paper.Firstly, hydantoin and its derivatives, which are widely used in organic synthesisand intermediates for the manufacture of pharmaceuticals and pesticides, werereviewed. Seven synthesis methods for hydantoin derivatives were also introduced.Secondly, eight hydantoin derivatives have been synthesized from the materialsof benzylamine and eight Boc-protected amino acids in three steps, which weredehydration, deprotection and cyclization. The overall yields of eight hydantoinderivatives were higher than 45%, and a practical synthetic method of hydantoinderivatives was developed.Lastly, 2-benzoxazolone, 2-benzimidazolone and 1,3-benzodioxol-2-one weresynthesized from 2-aminophenol, o-phenylenediamine and catechol, respectively. Thereactions were performed in mild conditions using triphosgene as carbonylation agentand triethylamine as base.Based on the mechanism of each reaction, the influence factors of the reactions,such as reaction temperature, reaction time, ratio of the materials, choice and quantityof the solvent, were detailed optimized in the paper. The workups of several reactionswere simplified. The optimal reaction conditions were obtained.
