Chondroitin sulfate is the basis of connective tissue of the mammalian, it mostly distributes cartilage, nose, throat, ligament and myolemma, especialy fish cartilage. CS is mostly used to treat neuralgia, arthritis, tinnitus, along with accelerating canker cicatrisation and preventing hyperlipemia . With lucubrating physiological action and biochemical character of CS, the effect of curor and preventing vein harden is also found. There is preferable curative effect for the preventing of coronary heart disease and myocardial infarction. CS can protect nerve cell, mucosal cell, kindey cell. CS is also executed toiletry mixed with something to protect cutaneous beauty.A method of dilute alkali-enzyme hydrolysis-ethanol precipitation to extract chondroitin sulfate from the sturgeon was studied. By making chief index on the glucosamine content and the yield of chondroitin sulfate, the best pick-up technics of CS has been found to offer important references on preparation and effective utilization for practice production.As CS band with protein, alkali extraction is propitious to destoty covalent bond among CS and protein to release CS. The process of alkali hydrolysis is the key of extracting CS. The process of emzyme hydrolyze is the main technology of improving production purity. Protein is one of the most impotant extraneous compontents of CS. Through emzyme hydrolyze, protein of the long chain among liquor and CS is more destoried and becomes soluble small molecule, such as peptide, amino acid. As it is difficult to reach the standard by using one kind of emzyme, the study uses two kinds of emzyme hydrolysis. Ethanol precipitation draws upon solubility difference of chemical constituents from cartilage to separate CS from other impurities.The result is as follows : the extraction with 2 multiple dosage of 6% NaOH solution at 40℃was churned up for 4h; Both pancreatin and pepsin were used to catalyze the hydrolysis of proteins in the fitrate : the dosage of pancreatin was 0.5% at pH 8.6 and 50℃for 3h, and then the dosage of pepsin was 0.3% at pH 5.7 and 40℃for 2h. The obtained hydrolysate was then removed by precipitation with 70% ethanol solution at pH 6.5 . The results showed that the glucosamine content was up to 27.13%, the yield of chondroitin sulfate was 43.27% and nitrogen content was 3.05%. The quality of products accord with the standard requirement of WSl-C3-0030-2000(Chondroitin sulfate tablets).
