Pumpkin is the fruit of Cucurbita moschata Duch which belongs to cucurbiaceae family. It has been used in food and medicine for thousands of years in China. The total output of pumpkin is 1.8 million tons in China, which is about 30% of world total output. This vegetable is very popular in China because it is thought to possess pharmaceutical effect such as lowering sugar and blood pressure. But there is little knowledge about the bioactive chemical constituents besides polysaccharides in pumpkin. In this study we focus on the chemical constituents of pumpkin.1. The chemical constituent in the chloroform extract in pumpkins was separated with silica gel column chromatography to yield 4 compounds: Cis-15-vaccenic acid (â… ), Steraric acid (â…¡), Cis-15-vaccenic acid methyl ester (â…¢) and Sugar (â…£), elucidated by MS and NMR data. These compounds were isolated from pumpkin for the first time. Cis-15-vaccenic (â… )Steraric acid (â…¡)Cis-15-vaccenic acid methyl ester(â…¢)Sugar (â…£)2. The n-butanol extract of pumpkins was isolated by D0402 macroporous adsorption resin eluted with water, 20%, 40%, 60%, 80% and 95% ethanol to obtain the components with different polarity. The fraction of 60% ethanol was isolated by HSCCC using a two-phase solvent system composed of light petroleum-chloroform-methanol-n-propanol-water(1: 5: 6: 1: 4, v/v) as the solvent system to yield three components BU1, BU2 and BU3. The component BU1 was isolated by HSCCC using a two-phase solvent system composed of light petroleum-chloroform-methanol-n-propanol and water (3: 5: 6: 1: 4, v/v) to yield a component BU1-2. The components BU1-2, BU2 and BU3 were isolated by preparative HPLC to yield compoundsâ…¤-â…¨respectively. Compoundsâ…¥-â…ªwere elucidated as moschglycolipid A-F, on the basis of ESI-MS, ESI-MS-MS and NMR analysis.
