The Eurotium cristatum is the advantage germ in the production of Fuzhuanbrick tea, it is the main factor that form the special quality of the Fuzhuan brick tea.In this research, the EC was separated from the Fuzhuan brick tea, then theextracellular polysaccharides were separated after 5day's liquid deep fermentation ofthe EC. The optimal condition for the extraction technique were gained. Then thecrude EC polysaccharide was purified by DEAE ion-extrange cellulose(DE-52)andSephadex G-100 gel filtrations and the separatiom curves were obtained. At last theMWs,forming monosaccharides of the purified EC polysaccharide were meatured.Theresults are as follows:1. Extraction Techniques of extracellular Polysaccharide of EC.The results showed that the optimal condition for the extraction technique were95% ethanol deposition for 10h as well as the ratiol:4. The contents of ECpolysaccharide were 62.2% and 70.1%.2. Isolation,purification and chemical structure of EC polysaccharide.ECP and ECP~# were isolated by DEAE ion-extrange cellulose(DE-52)andSephadex G-100 gel filtrations.GC and TLC determined the components of the saccharides.Their MWs weremeasured by Sephadex G-100 gel filtrations.The form of glucosidic bond wasdetermined by IR. Rha,Ara,Man,Glu,Gala were found in ECP-A1,ECP-A2,ECP-A~#1and ECP-A~#2 with the molar ratio of 12.73:2.47:23.75:1:1.79; 0.97:3.13:2.76:1:0.81;0.18:0.93:0.05:1:0.23 and 1.43:6.31:0.73:1:0.85, respectively.Their average MWswere estimated respectively to be 8.61×10~4D, 1.75×10~4D, 1.18×10~5D and2.6×10~4D.Typical absorption of polysaccharide was shown in their IR spectrum. Theresults of IR spectrum,Perodate oxidation and Smith reaction showed that the four polysaccharides were constituted by monosaccharide withα(1→3),α(1→6),α(1→2)orα(1→4) glucosidic bond.They had no typical absorption of nucleic acid or protein by UV scanning.ECP-A1 and ECP-A2 were first isolated from ECP-A;ECP-A~#1 and ECP-A~#2 werefirst isolated from ECP-A~#.
