Eurotium cristatum belongs to the genus Aspergillus glaucus, which is the preponderant fungi during the process of Fuzhuan tea flowering and plays an important role during the process of forming the unique quality of Fuzhuan tea.During the process of "flowering", Eurotium cristatum obtains the nutrients from tea,going on metabolism and transformation to meet its growth and development.Meanwhile, Eurotium cristatum produces various kinds of extracellular enzymes to promote a series of oxidation,polymerization, degradation and transformation in a variety of related substances in tea,which fulfil the material transformation of the unique color,aroma,taste and quality of Fuzhuan tea on the whole and make Fuzhuan tea have the unique function of health care.Polysaccharide is one of the four basic materials in composing life,mainly deriving from plants,animals and microbe.It is a class of important macromolecular compounds besides protein and nucleic acid and has a very wide range of application in the fields of the pharmaceutical industry,food industry and agriculture. Polysaccharide has minimal toxicity,in volves in all kinds of activities of the biological cell and has variety of biological function.In this paper,the main results are as follows:1.Using the phenol-sulfuric acid to determine the polysaccharide content in Eurotium cristatum.By the test, the determination of the optimal conditions is decided:6%of phenal with1mL,concentrated sulfuric acid with5mL and100℃of water bath with15min.Under the optimum conditions,the standard curve equation is y=0.0132x+0.0225and the coefficient of linear correlation is R2=0.9968.The test determines the polysaccharide content in Eurotium cristatum is2.76%.2.Adopting the method of the water extraction and alcohol precipitation to extract the crude polysaccharide from Eurotium cristatum.By the single factor test and the selection of orthogonal test condition,it determines the optimum conditions of polysaccharide in Eurotium cristatum:the temperature of extractive polysaccharide is 95℃. It is extracted by3times and takes60min. And the ratio of solid-liquid is1:20.3.The wall-breaking technology of Eurotium cristatum ascopore is researched respectively by the methods of mechanism,chemistry and biology. The results of test show that immersed in distilled water-temperature difference-vibration is the best wall-breaking method of Eurotium cristatum ascospore and the rate of wall-breaking is90.8%.It has determined the polysaccharide content in Eurotium cristatum which is wall-broken and the rate of the polysaccharide content is4.28%,which shows that wall-breaking has positive significance for nutrients releasing in Eurotium cristatum.4.Use of liquid chromatography method for the determination of Eurotium cristatum in monosaccharide composition of polysaccharide and analysis of the monosaccharide content. The Eurotium cristatum polysaccharide composed by mannose, rhamnose, Arabia sugar, glucose and galactose. Through the spectral analysis and standard curve calculation, the content of monosaccharide in Eurotium cristatum is mannose:rhamnose:Arabia sugar.glucose:galactose=42.4%23.08%:8.68%:12.2%:13.6%.
