The relationship between smoking and health has being regarded for a long time. Cigarette smoke is a mixture of about 4000 compounds, a lot of which are harmful to human health. Mainstream cigarette smoke, because of its direct contact with smokers, has being studied fervidly since 1960s. On the basis of former studies, the analysis methods and their application of several kinds of deleterious compounds, including 4 kinds of tobacco-specific nitrosamines and 7 kinds of phenolic compounds in mainstream cigarette smoke, have been studied in this paper. Our research work for the deleterious compounds involved the following contents and achievements:1. A method has been developed for simultaneous determination of four major tobacco-specific nitrosamines (TSNAs) in the particulate phase of mainstream cigarette smoke by high performance liquid chromatography-electrospray ionization-mass spectrometry (HPLC-ESI-MS/MS). Mainstream cigarette smoke particulates, produced using standardized machine smoking protocols, were collected on a 44 mm Cambridge filter pad. The particulate matter was extracted using 0.1 mol/L ammonium acetate, purified by solid-phase extraction, and analyzed by HPLC-ESI-MS/MS. The limits of detection of each TSNA ranged from 0.04 to 0.07 ng/mL; the recoveries ranged from 94.88% to 100.15% and the coefficient of variability is below 7.3%. This method showed high sensitivity, specificity, throughput and no evidence of artifact formation of TSNAs. Some brands of cigarettes were analyzed and showed good results.2. A method has been developed for simultaneous determination of seven kinds of phenolic compounds in the particulate phase of mainstream cigarette smoke by high performance liquid chromatography. Mainstream cigarette smoke particulates produced using standardized machine smoking protocols were collected on a 44mm Cambridge filter pad. The particulate matter was extracted using 1% (V/V) acetic acid, filtered by 0.45μm filtration membrane, and analyzed by high performance liquid chromatography (HPLC). The limits of detection of the phenolic compounds are all below 0.1μg/mL; the coefficient of variability is below 5% and the recoveris range from 94.7% to 99.9%. This method has high sensitivity, simple operation and good stability.3. Applying the method studied this work, four major tobacco-specific nitrosamines and seven kinds of phenolic compounds in 28 brands of cigarettes, including domestic and overseas, were analyzed. The distributing rules of these deleterious compounds at different brands of cigarettes were found by analyzing the data and the feasibility of some of deleterious compounds reducing method in mainstream cigarette smoke was studied.
