Oviductus Ranae is the dry oviduct of feamale Rana temporaria chensinensis, which is a kind ofwell-known medicinal materials in northeast area of China.It is not only benefit for your kidney, lung andso on but also has high officinal and nutritious value. Oviductus Ranae is rich in protein. Crude protein is40 percent. The amount of amino acid is 17.6 percent, essential amino acid is 12.8 percent. Meanwhile, thekinds of amino acid is complete and the component of amino acid is reasonable, it is ideal nutrition proteinorigin to humanity. Oviductus Ranae also contains much vitamin and microelement such as zinc,manganese and copper. It also has much physiological activator, such as estradiol, progestin andtestosterone and so on. It is not only high-grade nutrition and herb, but also is a kind of important goods forexports of officinal animal in china.Hydrolysis of animal protein is good flavors, health and nutritious but also tended to absorption byhuman body. it wins sb.'s high favour in food industry. The protein is the main nutriment that the humanbody needs, it is absorbed in the form of amino acid peptide in human body. The modem researchdiscovered that small peptide of non-sole amino acid has importantly physiological function toabsorption metabolism in human body. it is superior to sole amino acid in digestive absorption,and isdifferent internal transport from sole amino acid.The research use Oviductus Ranae as main material and trypsinase,papain, animal protease, neutralprotease, bromelin as proteolytic enzyme; Optimize the condition of proteolysis through study on conditionof temperature,time, substrate concentration, enzyme dose; prove up component and nature of protolysate.developed nutrient medium of Oviductus Ranae bioactive polypeptide and amino acid that is rich innutrition. The research result showed as follows:1. The optimal hydrolysis condition with trypsinase was as follows: pH 8.5, hydrolysis time 6h,E/S 36percent, hydrolysis temperature 45℃,substrate concentration 1:30.the degree of hydrolysis 59.83 percent,extraction yield of peptide 47.73 percent, solubility 52.63 percent.2. The optimal hydrolysis condition with papain was as follows: pH 7.0, hydrolysis time 6h,E/S 32 percent,hydrolysis temperature 60℃,substrate concentration 1:30.the degree of hydrolysis 35.01 percent,extraction yield of peptide 39.46 percent, solubility 44.12 percent.3. The optimal hydrolysis condition with trypsinase and papain was as follows: first trypsinase and second papain. The ratio of additive enzyme (trypsinase: papain) 2:1,total quantity 36 percent,the ratio of time2:1,total time 6h,substrate concentration 1:30. the degree of hydrolysis 62.13 percent, extraction yield ofpeptide 55.99 percent, solubility 61.53 percent.4.Trypsinase and papain fractional hydrolysis was determined as optimal hydrolysis condition bycomparision.5.High solubility was showed in TCA-NSI of hydrolysate with trypsinase and papain with pH2-10;viscocity reduced clearly comparing with substrate protein;molecular weight was 25KDa-15KDa,15KDao8KDa, 8KDa-2KDa, the kind of amino acid was complete.6.Anion exchange resin and cation resin was used to desalt. The rate of desalination 90.05 percent, nitrogenrecovery rate 92.05%,and showed certain discolouration.7.The formulation of nutrient medium of klinitamin was as follows:β-cyclodextrine quantity 2.5percent,crystal sugar quantity 4 percent, Honey quantity 6 percent. Bactericidal condition 105℃, 20min.
