Bilayer lipid membrane (BLM) is a reconstituted biomembrane in vitro. It mimics the configuration of biological cell membrane. The bilayer configuration of BLM facilitates modifier embedding. So BLM has been considered as a good model of the membrane system for studying of life phenomenon. The type and quantity of modifiers embedding into BLM can be well controlled. By embedding a modifier (such as a kind of antibody, or enzyme, etc) to BLM, it can peculiarly recognize the counterparts like cell membranes, which have the function of the extraordinary identification. So BLM modified can be used to identify and detect the counterpart in solution. As An ideal sensitive material of biosensors, BLM will be widely explored for the development of novel biosensors sensing to various species.Electrochemical measurement method is a common traditional way to study the forming process of BLM. It can character the overlay of BLM in an exiguity area. However, the applied voltage during measuring may affect the performance of BLM. This research adopted a new method, namely surface plasma resonance (SPR) technology which is an optical method, to investigate the characteristics of supported BLM. Then, the combination of electrochemical measurement method and SPR was applied for BLM investigating and analyzing.Based on the theory of SPR, a simple mini-SPR system, which accuracy is suit for the present investigation, was constructed for BLM studying. The particular characteristic of the system is that the prism, one of the key parts of the system, can be replaced conveniently. The system benefits to the contrast experiments on BLM investigation for the study and exploration of the basic properties of BLM. Also, it can be utilized to analyze the effect of electrochemistry methods on BLM.The metal substrate was treated by mechanical methods. Then, the surface processed was activated with some surfactant. By this way, it can improve the consistency of microstructure of substrate. Also, it can ameliorate the comparability and repeatability of experimental datum. Some assistant experiment tools, such as setup for probes fabrication, sharpener, micro-adjusting platform, the flowing cell and so on were prepared. All of these contribute to the experimental performance. For BLM formed well, the effect of the compositions of the buffer solution and phospholipid solution was studied. Based on the experimental results, the optimum ration of ingredients of BLM forming solution was obtained.The principle that phospholipid molecules can self-assemble on the nascent metal surface was analyzed based on the metal bond theory and atomic energy theory. In our opinions, BLM self-assembled could be the course of rupturing and rebuilding of metal bonds, and transporting of the atomic energy from high-energy state to lower state.
