In recent years,polysaccharide has attract much attentions for its extensive biologic character. Acid mucopolysaccharides have immense applied value in foodstuff,medicament,cosmetic and so on;so it is practically significant to research and determine acid mucopolysaccharides.Resonance Rayleigh scattering(RRS)technology has high sensitivity and simple apparatus,and has arose the researcher's extensive attention.It has been developed quickly in recent years and has been applied in the determination of biomacromolecules,trace metal ions and nonmetals as well as organic matters.In this paper,we have determined three type of acid mucopolysaccharides(sodium hyaluronate,dextran sulfate sodium,heparin)with the probes of crystal violet,diantipyrylmethane, gold nanoparticles by the RRS technology,we have proposed the triple-wavelength overlapping resonance Rayleigh scattering method,studied its applications concretely and discussed the reaction mechanism.The main contents are as follows:1.Determination of sodium hyaluronate with crystal violet by resonance Rayleigh scatteringIn pH 4.2 acidic medium,the resonance Rayleigh scattering intensity of hyaluronate(SH)or crystal violet(CV)is very weak while it will enhance greatly when CV and SH form an ion-association complex,which results in a significant RRS enhancement of the system.The maximum scattering wavelength appears at 715 nm.We have investigated the RRS spectral characteristics and the optimum conditions of the system and founded a fast,precise method to determinate the SH.The linear regression equation obtained isΔI= 202.1 + 206.8C(μg/mL)and the linear range is 0.2~40μg/mL,the detection limit(3δ)is 15 ng/mL.This method can be used for the determination of total amounts of SH in samples with satisfactory results.2.Determination of dextran sulfate sodium with crystal violet by triple-wavelength overlapping resonance Rayleigh scatteringA triple-wavelength overlapping resonance Rayleigh scattering(TWO-RRS)method was developed to detect dextran sulfate sodium(DSS)with CV.At pH 10 BR buffer,the interaction of CV with DSS occurred which greatly enhanced the RRS intensity with the new RRS peaks appearing at 340,501 and 671 nm and all these three peaks enhanced with the increase of DSS concentration in the range of 0.04~2.5μg/mL and the detection limit for the three single peaks was 24,27,and 27 ng/mL,respectively,whereas that of the TWO-RRS method was 13 ng/mL.The TWO-RRS method was found to have much better flexibility and higher sensitivity than traditional single-wavelength method.3.Resonance Rayleigh scattering method for the determination of dextran sulfate sodium with diantipyrylmethaneA new method for the determination of dextran sulfate sodium(DSS)with diantipyrylmethane (DAM)by RRS technique has been proposed.In pH 10 BR buffer,the interaction of DSS with DAM occurred which greatly enhanced the RRS intensity that locat at 352 nm.The interaction conditions and affecting factors have been studied.Under the optimum condition,the enhancement of the RRS signal is directly proportional to the concentration of DSS in the range of 0.05~2.5μg/mL and its detection limit is 17 ng/mL.4.Determination of dextran sulfate sodium with gold nanoparticles by resonance Rayleigh scattering techniqueWe have determinated the dextran sulfate sodium(DSS)by the resonance Rayleigh scattering technique(RRS)with the new probe of gold nanoparticles(GNP).In the HAc-NaAc buffer GNP interacts with DSS which could result in the the remarkable enhancement of the RRS intensity and appearance of new RRS spectra that located at 332 nm.The conditions of the reaction and influencing factors were investigated.Under the optimal conditions the enhanced RRS intensity is in proportion to the concentration of DSS in the range of 0.01~2.5μg/mL and its detection limit(3δ) is 1.85 ng/mL.5.Determination ofheparin with gold nanoparticles by resonance Rayleigh scattering technique A new resonance Rayleigh scattering method for determining heparin(Hep)with gold nanoparticles(GNP)is presented.In pH 4.7 HAc-NaAc buffer solution,it is found that when the GNP and Hep react through electrostatic and hydrophobic forces to form the complex(GNP-Hep) quickly,the RRS intensity of the system can be greatly enhanced.Based on this phenomenon,a sensitive and fast method for determination of heparin was established.This method is satisfying when applied in the determination of actual sample.
