Food safety has been turned to the focus of society and government, and attracts people's attention. Therefore, to strengthen people's ideas about dietary supplement, to recognize adulteration of synthetic drugs in dietary supplements correctly and to provide scientific and efficacious analytical methods, these have important realistic meanings. Based on high sensitivity, strong specificity, high reliability and simple pretreatment of high performance liquid chromatography-mass spectrometry (HPLC-MS), the synthetic drugs in dietary supplements and traditional Chinese medicines for weight reducing or diabetes mellitus were determined, and a new method for safety monitoring of dietary supplements-quality profile was proposed, and the main contents as follows:1. The status of adulteration and detection of synthetic drugs in dietary supplements, and the common methods for detection of synthetic drugs had been summarized. The advantages of HPLC-MS in detection of synthetic drugs had been discussed.2. An analytical method was developed for the simultaneous determination of fenfluramine, phenolphthalein, N-di-desmethyl sibutramine, N-mono-desmethyl sibutramine, sibutramine, and orlistat in herbal weight reducing dietary supplements by HPLC-electrospray ionization (ESI)-MS. Separation of HPLC and response of ESI-MS were optimized, and ion suppression of targets in sample matrices was investigated. The method has the advantages of low detection limit, high reliability, and it can detect the six synthetic adulterants in herbal weight reducing dietary supplements accurately. It provides the reference for departments of quality inspections.3. A HPLC-ESI-MS method for simultaneous determination of phenformin, rosiglitazone, glibenclamide and glimepiride in dietary supplements and traditional Chinese medicines for diabetes mellitus was proposed. We optimized separation of HPLC, response of MS, and determined the four targets with qualitative and quantiative results. The method was applied for detecting the four illegal additives successfully.4. A new method for safety monitoring of natural dietary supplements-quality profile was proposed. It would convert passive monitoring of synthetic drugs to active, guarantee security of dietary supplements, and prevent addition of synthetic drugs after "approval". HPLC and ESI-MS were applied in quality profile, and it is proven that quality profile used in monitoring illegal additives is feasible.
