Probiotics have some functionality, and long producing history in foodstuff industry either. While probiotics are frail to lead to no long time preservation, they can't tolerate gastric acid to get to the intestine. Probiotics get containment and don't field planting in intestinal tract to educe physiological functions because of nutritional deficiency and cholate corrosion and so on. Fungus encourage multiplication and retention for probiotics when they are used as a kind of prebiotics in microcapsule. The experiment took three aspect of researchs:1. Studied the best extracting technology of fungus polysaccharidess and the effect of polysaccharidess on probiotics growth to search somatomedin of probiotics. Method to extract polysaccharidess was alkalinous extraction and alcohol sedimentation and compared effect of different kinds of fungus on probiotics growth. Separated polysaccharidess with DEAE-cellulose column chromatography, analyzed their purity quotient with Sepharose CL-4B gel column chromatography and get three main hump that polysaccharidessⅠ,polysaccharidessⅡ-c and polysaccharidessⅢ-b. Determined growth curve,growth OD and pH to compare differences amone probiotics growth when adding 1% three kinds of polysaccharidess into MRS medium.The best extracting technology for domestic fungus polysaccharidess was 100℃,5h,pH=10 and 5 multiple 95% alcohol sedimentation. There were not too much different effects of domestic fungus polysaccharidess on probiotics growth. PolysaccharidessⅠand polysaccharidessⅡ-c promoted growth for probiotics obviously and produced lactic acid for Lactobacillus acidophilus, while polysaccharidessⅢ-b got secondary multiplication effect because of high content of giant molecule polysaccharidess and protein-polysaccharidess compounds. Probiotics got 1010cfu/ml viable organism concentration or more when they were during log phase. Fungus powers promoted OD growth and produced lactic acid for Lactobacillus acidophilus obviously.2. Took Sodium Alginate,Gelatinum-Gum Arabic and Flaxseed gums as paries metrial to research effect on some important technical parameters of microcapsules. Prepared Sodium Alginate and Gelatinum-Gum Arabic microcapsules as contrasts to investigate effect of flaxseed gums on microcapsules efficiency and constitution with electron microscope.0.5% concentration flaxseed gums were added into microcapsules on base of Sodium Alginate, and got 96% envelopment efficiency; There were not too much effect of the first agitating on encapsulating efficiency but much on envelopment efficiency oppositely. Taking 20min×700rpm at the first agitating could get 90% or more envelopment efficiency; There were much effect of the second agitating on encapsulating and envelopment efficiency. Taking 15min×400rpm at the second agitating could get high encapsulating and envelopment efficiency at the same time; The microcapsules that added flaxseed gums with more smoothed surface and more compact texture than that added Sodium Alginate and Gelatinum-Gum Arabic. Microcapsules that added flaxseed gums had good intestines dissolubility, acid resistance, cholate resistance and high temperature resistance, and got complete disaggregation at 60min in artidicial intestinal juice with a 90.00% envelopment efficiency that was 15.92% higher than Gelatinum-Gum Arabic microcapsules; There was a 80% or more survival rate for microcapsulation probiotics when stayed in artificial gastric juice for 2h, 73% survival rate in 0.5% cholate for 12h, and 70% survival rate in 0.5% cholate for 12h; Dealing with 60℃×60min got 44% survival rate that was 9% higher than the other two kinds of microcapsules.3. Added domestic fungus as prebiotics into microcapsules, and prepared synbiotics microcapsules.Synbiotics microcapsules had one more order of magnitude than the microcapsules that without fungus when dealed with artificial intestinal juice for 12h; 8% fungus addition was good; Prociotics microcapsules had depressant effect on enteropathogenic E.col, Salmonells enteritidis and Shigella sonnei when they were digested by artificial intestinal juice, especially Lactobacillus casei and Lactobacillus plantarum microcapsules with bacteriostasis diameter that 20.72mm,19.98mm,16.10mm and 19.64mm,19.90mm,18.64mm; Determined probiotics survival rate at the definite time and calculated holding time on the basis of micro-organism inactivation dynamics formula, there was a result that the experimental microcapsules would be held for 172 days at 4℃.
