High performance Capillary Electrophoresis(HPCE)is a novel analytical technique developed in 1980 for the separation of compounds based on the differences in the migration velocity of ions or solutes through the given medium in the electric field.Due to its high separation efficiency, high sensitivity,fast separation,small sample required,low consumption of chemicals and a diverse range of application,HPCE has become one of the most important techniques for the analysis of medicine and Chinses medicine.In this thesis,the principle and characteristic of HPCE and its applications in analysis of medicine were introduced.The main aspects are showed as following:1.Separation of Shikimic Acid by High Performance Capillary Electrophoresis.Effects of several important factors were investigated such as the pH and concentration of running buffer,operation voltage,injection time and additive.Shikimic acid can be separated under the optimized conditions:30 mmol/L Tris+30 mmol/L H3BO3(pH=8.6)as running buffer,15 kV operation voltage,10 s as injection time.2.A quick and simple method of High Performance Capillary Electrophoresis with conductivity detection was established for the detection of Cimetidine.Separation was carried out in a uncoated fused silica capillary (50 cm×75μm).Effects of several important factors were investigated such as the pH and concentration of running buffer,operation voltage,injection time, injection height.Cimetidine can be detected in nine minutes and forty seconds under the optimized condition:30 mmol/L Tris+100 mmol/L Cit(pH=8.26) as running buffer,10 kV operation voltage.The concentration of Cimetidine was lined with the peak height in the range of 0.5~20 mg/L, h=9.0923c+15.83,r=0.9984.Recovery rate between 98%—108%.3.With alkali water as the solvent,flavone can be extract from Phyllanthus urinaria L.by means of ultrasonic waves.The effect is compared with traditional ethanol extraction.The results demonstrate higher extraction rate and efficiency by ultrasonic waves.The optimum conditions were identified as the follows:pH of Ca(OH)2 was 11,the time of ultrasonic tratment was 40min,the ratio of solid to liquid was 1/50.The extraction rate was 1.95%under the optimum conditions.4.A method of High Performance Capillary Electrophoresis was established for the separation of flavone in Phyllanthus urinaria L,the concentration of rutin in Phyllanthus urinaria L was detected.Effects of several important factors were investigated such as the pH and concentration of running buffer,concentration of SDS,operation voltage,and injection time. Under the optimized condition:60 mmol/L Na2BO4+16 mmol DS(pH9.0)as running buffer,15 kV operation voltage,the detection wavelength was 245 nm,the concentration of rutin was lined with the peak height in the range of 20~500 mg/L(r=0.9989),the average recovery rate between 93.2%—105%, RSD lower than 8%.
