Nimodipine (NMD) is a calcium antagonistic dihydropyridine derivative with a preferential effect on cerebral vessels. Its effect on the peripheral blood vessels is less pronunced so that NMD increases cerebral blood flow without substantial decrease of blood pressure. Sales volume of NMD is rising gradually in china. Based on the quality problems of the nimodipine product made in China, such as mixed polymorph and asymmetry CSD, the crystallization process and the polymorphism were investigated and analyzed systematically.Firstly, two different polymorphic forms of NMD were prepared. Infrared absorption spectrum, Thermal analysis, Powder X-ray diffraction were used to analyze these crystals. Secondly, it was studied on the solubility and dissolution of two different crystal forms. The results showed polymorphism effects drug's dissolution characteristicsAimed at the quality demand of the nimodipine crystal product, the effects of operational parameters such as crystallization method, cooling rate, agitation intensity, and seed etc. on the CSD, yield and morphology were investigated systematically by experiment and the experimental optimum operational conditions for the crystallization of nimodipine were finally obtained. The work about the crystallization new technique for nimodipine, which was done in this paper, has not been reported in any literatures.
