In this paper, extract of Sanguisorba officinalis was tested for its antioxidant activity, including investigation of its scavenging effects on three kinds free radicals. The processing conditions of leaching active substances from Sanguisorba officinalis were optimized with the orthogonal design test. The sensitivity to universal factors of the extract was studied. At the same time, the isolation and purification of the antioxidative components of Sanguisorba officinalis were also studied .The optimum extraction conditions of every factor for leaching active substances determined by the single factor experiment method were as follows: the concentration of ethanol was 60%; the ratio of material and solvent was l:12, temperature for extracting was 70°C with 2h. With the orthogonal design test, the results showed that the optimum extracting condition was: the concentration of ethanol was 60%; the ratio of material and solvent was l:12, temperature for extracting was 73.1°C with 2.09h. Under this condition, the content of tatal flavinoids of the extract was 0.2104mg/mg, the content of tatal phenols of the extract was 0.1938mg/mg.The stability to temperature and ultraviolet of the extract of Sanguisorba officinalis were also studied. The result showed the extract was very stable to temperature and ultraviolet.All extracts obtained in different fractions by using different polarities of solvents such as petroleum ether, chloroform, ethyl acetate, normal butyl alcohol and water were compared through the testing of their antioxidative activities.The scavenging effects on superoxide anion free radical(O2ˉ·) and hydroxyl free radical(·OH) of every extraction were studied by three photometric methods , BHT was the reference . The results showed that every extraction except the petroleum ether of the extracts from the Chinese traditional drug Sanguisorba officinalis had capacity on scavenging·OH free radical , and had no obvious capacity on scavenging O2ˉ·free radical .The results showed the extraction of ethyl acetate had a strong capacity on scavenging·OH free radical and normal butyl alcohol took second place. To track antioxidant components in the extracts of ethyl acetate in which many different antioxidant points appeared after diffusion. The qualitative analysis of extracting sample is conducted by the specific reaction of total flavonoids with AlCl3 and so on, It is confirmed that the extraction material is flavonoids.The extracts of ethyl acetate of Sanguisorba officinalis was seperated by polyamide columnchromatography. The antioxidant component was determined by LC-MS. The results showed that one of antioxidant components was kaempferpl-3,7-dirhamnoside.The extract of Sanguisorba officinalis was applied in cosmetics. All the results confirmed that extract of Sanguisorba officinalis has the antioxidative effect, the skin-lightening effect and the antimicrobial effect. So it can be widely used in cosmetics.
