Plant polyphenols, an excellent natural antioxidant, has been studied extensively, while the extracting techniques of polyphenols, oligoprocyanidms and flavones in the root of sanguisorba haven't been reported in China.It was investigated how to increase the yield and purity of tannin, flavone and oligoprocyanidms from the root and stalk of sanguisorba, and the stability of polyphenols from sanguisorba was studied, too. The chemical constitutes of sanguisorba were analyzed, the conditions on extracting tannin, flavone and oligoprocyanidins from sanguisorba by general lixiviating process and ultrasonic wave were optimize. The flavone and oligoprocyanidins were separated from sanguisorba and purified. The stability of polyphenol from sanguisorba was studied and the chemical structure of the products was analyzed. The results are as follows:The root of sanguisorba is abundant in starch (16.1%), tannis (8.85%), oligoprocyanidins (5.7%) and flavone (14.56% ). The content in root is higher not only than stalk and leaves, but also than the normal plants.In lixiviating process, the preferable technical parameters of extracting tannis, flavone and oligoprocyanidins from the root and stalk of sanguisorba were obtained through changing the concentration of ethanol, particle size of raw material, the ratio of sample weight to solvent volume, the reflux temperature, extracting time and reflux times. The extraction rates of tannin, flavone and oligoprocyanidins from the root were 8.53%, 14.56% and 5.7% respectively (to the dry raw material), and in the stalk were 2.32%, 0.90%and 2.08% respectively.In the extracting process using ultrasonic wave, the preferable technical parameters of extracting polyphenols from the root and stalk of sanguisorba were acquired by changing the particle size of raw material, the ratio of sample weight to solvent volume, ultrasonic time, ultrasonic power, ultrasonic times. The extraction rates of polyphenols in the root and stalk were 11.71% and 1.88% individually.The flavone and oligoprocyanidins were purified by the solvent separation method. The proper separation condition was obtained. In the obtained products, the contents of total flavone and oligoprocyanidins were 96.79% and 56.85%, and the yields were 7.59% and 3.42% respectively.The stabilities of the extract from sanguisorba under different pH, light, heat, ultraviolet ray, oxidant, reducing agent and additive agents were studied. It was indicated the products could widely used in the condition of weak acid or neutral, while they were not stable to heat and light, so they should be used without light and at low temperature. The products had a strong ability to withstand ultraviolet ray. They could easily be oxidized and degraded by oxidant while they were not easily affected by reducing agent and were almost not affected by additive agents. The extract from the stem of sanguisorba was analyzed by UV and FTIR, showing that the structures of the active components were not changed during ultrasonic waveextracting.This research offers a potent basis for the exploitation and utilization of sanguisorba in medicine, health-care food and cosmetic.
