Inecstigators pay more attention to the problems of environmental security along with widely used of chiral pesticides.In this paper,a method of chiral separation of organic Phosphorus pesticid: O,O-dimethyl-1-(3-trifluoromrthylphenoxyacetoxy)ethylphosphonate(Peng5) and O,O-dimethyl-1-(4-methyloxyphenoxyacetoxy)ethylphosphonate(Mo5) was developed by high performance liquid chromatography(HPLC).Daphnia magna was used as test acute toxicity of enantiomer,and also provides the evaluation standard for environmental organisms.A method of chiral separation of Mo5 and Peng5 was developed by HPLC on the Chiralpak AD.The effect of chromatographic conditions such as alcohols modifier in the mobile phases,added amounts of isopropanol in the mobile phases,column temperature to recognition of the Peng5 and Mo5 were studied,the thermodynamical mechanism of enantioseparation have also been preliminary discussed.A simultaneous baseline separation of the enantiomers was obtained with a mobile phase of n-hexane/isopropanol (90:10(Mo5)/95:5(Peng5) at a flow rate of 1.0mL/min and wave length 220nm under 25℃in Chiralpak AD-H column.As the main aquatic organism,the acute aquatic toxicity of Peng5 and Mo5 to Daphnia magna were investigated.According to OECD method,the 24h-LC50 and 48h-LC50 were adopted to evaluate the acute aquatic toxicities of the compounds.the results showed that 24h-LC50 and 48h-LC50 values of Rac-Peng5,Peng5-pk1,Peng5-pk2 for Daphnia magna in the experimental conditions were 9.41mg/L,3.70mg/L,5.79mg/L and 1.29mg/L,0.61mg/L,0.69mg/L respectively.And the toxicity order was:Peng5-pk2>Peng5-pk1>Rac-Peng5;24h-LC50 and 48h-LC50 values of Rac-Mo5,Mo5-pk1,Mo5-pk2 for Daphnia magna in the experimental conditions were 22.77mg/L,9.58mg/L,7.24mg/L and 0.39mg/L,0.32mg/L,0.30mg/L respectively.And the toxicity order was:Mo5-pk2>Mo5pk1>Rac-Mo5.
