In order to fully exploiture the Oil camellia resources,provide the technical support for its comprehensive utilization,to enhance the economic efficiency of Oil camellia industry,and promote Oil camellia industry profession development,I did many hard work,my research was focused on the extraction of xylose from Oil camellia shell by utilizing of acid hydrolysis and fermentation,The main content is as follows:1.Analyses of the principle components in Oil camellia shell:The principle components of Oil camellia shell,such as cellulose,xylogen,hemicellulose,protein,fat,and tea saponin were determined with chemical methods.The content is 23.45%,21.56%,9.70%,3.24%,3.69%, 5.17%,respectively.2.Comparation of xylose assay methods:By spectrophotometry,the quantitative determination of xylose with the method of Douglas color reaction was compared with the method of DNS color reaction.It was shown that the former had higher absorbency and was more sensitive than the same sample.It was also found that,even if there is some hexose in sample,the result would be no difference between two methods.For detectin fermentation type xylose content with Douglas color reaction,the peak absorption wavelength was 552 nm,the standard regression equation was Y=2.076X+0.0994 with the correlation coefficient r=0.9994,standard deviation 2.17%,and standard error 0.79%.The recovery rate of adding standard sample was 94.60%with the relative standard deviation 1.73%.3.Screening technological condition(parameters) of Acid Hydrolysis for extracting Xylose from Oil camellia shell:The orthogonal test design was adopted to study the optimal parameters of Hydrolysis.The results showed that the raw optimum condition for the hydrolysis technique is as follows:the hydrolytic temperature is 120℃,material/solvent ratio is 1:5,the time for hydrolysis is 2 hours.Sulfuric acid concentration is 2%.By using the optimized condition;the extracting rate can reach 9.11%.4.Study on the extraction of Xylose from Oil camellia shell by Fermentation White-rot fungus utilized in this study is provided by our microbiology laboratory.The composition of liquid medium optimized by the response surface method is as follows:Oil camellia shell 4%(NH4)2SO4 1%MgSO4.7H2O 0.6%.By using the optimized medium,the extraction rate reached 2.89%. Through shake-flask fermentation trials,the culture conditions were found to be as follows:the initial pH of medium is 5.5,the medium volume is 100 mL/250 mL,the inoculation amount is 10%,culture temperature is 30℃,culture time is 48 hours,the revolution is 120r/min.Fermentation under this condition,the extraction rate reached 2.96%.
