Functional magnetic particles are widely used in bioseparation, targeted drug,immunoassay,and other fields because of their abundant active functional groups(-CHO,-OH,-NH2 and-COOH) and their magnetism.Comparing to the traditional separation technology,the separation method based on the functional magnetic particles is high efficient,fast,and environment-friendly.In this paper,the preparation of the functional magnetic particles and its application on the separation and extraction of alkaloids in Chinese herbal medicine,as well as drug delivery,are studied.The details are summarized as follows:1.The synthesis method of the functionalized magnetic particles and its application in the bioseparation,the various extraction methods on separating and extracting alkaloids in Chinese herbal medicine and the comparison of their advantages and disadvantages,were reviewed.2.Polystyrene-acrylic acid magnetic particles(p(St-AA) MPs) were prepared using microemulsion polymerization and the prepared p (St-AA) MPs material was characterized.The amount of magnetic fluid,styrene,acrylic acid,and the kind of the initiators were optimized.A simple,economical method for synthesizing p(St-AA) MPs with rich surface functional groups and good magnetic responsiveness was established. 3.The alkaloids in tea and Coptis chinensis franch were separated and extracted with p(St-AA) MPs as the carrier and high-performance liquid chromatography(HPLC) as the analytical method.A novel method for separating and extracting alkaloids in Chinese herbal medicine without organic solvent was developed.The separation conditions,such as the pH in the feed solution,the extraction time,the kind and the composition of the back-extraction solution,were optimized.4.In order to obtain non-toxic,biodegradable functionalized magnetic particles,carboxymethyl chitosan magnetic nanoparticles (CMCS-OA-Fe3O4 NPs)were prepared by a two-step method,i.e., synthesizing nanosized Fe3O4 in the presence of oleic acid(OA-Fe3O4 NPs),then directly coating the OA-Fe3O4 NPs with carboxymethyl chitosan in aqueous solution and the prepared CMCS-OA-Fe3O4 NPs material was characterized.The conditions for synthesizing CMCS-OA-Fe3O4 NPs were optimized.5.The alkaloids in Coptis chinensis franch were separated and extracted with CMCS-OA-Fe3O4 NPs as the carrier and high-performance liquid chromatography(HPLC) as the analytical method.The separation conditions were optimized.The drug loading and the sustained-release property of the CMCS-OA-Fe3O4 NPs for adriamycin(ADM) in phosphate buffer solution(PBS,pH 7.4) were studied.The results indicate that CMCS-OA-Fe3O4 NPs may be used as the magnetic-targeted nano drug carrier for the treatment of tumour.
