Soybean lecithin contains primarily PC, PE, PI, SM, LPC and PA. The fractionation of these phospholipids is desirable for certain applications. Among them, phosphatidylcholine (PC), which has special character and physiological function, is widely used in food, cosmetic and pharmaceutical industries. But almost all of the high purity PC consumed in our country being abundant in soybean resource is imported. Thus there is a demand for the technology of preparation for PC with high quality.In order to improve the content of PC in lecithin, a study has been carried out to obtain de-oiled powder phospholipids from soybean oil residue using acetone. Acetone insoluble residue(AI)was used as the indicator to show the content of phospholipids. The effects of extraction temperature, extraction time, the ratio of soybean oil residue to acetone, extraction times were evaluated. We got the best result when extraction time was 20 minutes, extraction twice, extraction temperature was 40℃, the ratio of soybean oil residue to actone was 1:20(g:mL), and the acetone insoluble residue (AI) was up to more than 96% compared with soybean oil residue was 14%.Sodium carbonate solution and calcium chloride solution was used to treat soybean oil residue, TLC was used for qualitative analysis. It was found that LPC and SM were almost removed when product was treated by 50v% ethanol solution.Powder lecithin was treated by ethanol solution. Optimum condition was determined to get purified PC enrichment. The effects of temperature, concentration of ethanol solution, extraction time, the ratio of soybean phospholipids powder to the solution of ethanol, extraction times on the yield and PC content were evaluated. Optimum technological parameters were obtained as follows: temperature was 35℃, concentration of ethanol was 95%, extraction time was 30 minutes, the ratio of soybean powder lecithin to alcohol was 1:5(g:mL), extract twice. The content of PC has been improved from 34% to more than 52%.
