In present study, potato protein was utilized as the raw material to investigate the enzymatic hydrolyzation by using different enzymes.The optimal protease was choosed by the degree of hydrolysis, dissolved free amino acids and bitter value.Potato polypeptides can be gained by hydrolyzing potato protein.Different enzymes would have different degree of hydrolysis. Primary, choosed six enzymes: the 2709 alkalescence protease,the bromelain,the plant protease,the Alcase protease,the Neutrase protease,the Flourzyme protease.In this thesis, the best condition of hydrolyze by the investigation of the hydrolysis time;the temperature of the reaction; the substrate concentration;the ratio of enezyme and substrate;pH.The results as follows:The 2709 alkalescence protease: the ratio of enezyme and substrate:0.04%,pH:8.0,the temperature of the reaction: 50℃,the substrate concentration:11% ,the hydrolysis time:2h. The best condition of the degree of hydrolysis can be come to 30.94%.The bromelain: the ratio of enezyme and substrate: 0.8%,pH: 6.0,the temperature of the reaction:50℃,the substrate concentration:8%,the hydrolysis time:4h.The best condition of the degree of hydrolysis can be come to 26.00%.The plant protease: the ratio of enezyme and substrate:0.3%,pH:6.0,the temperature of the reaction:50℃, the substrate concentration:9%,the hydrolysis time:4h.The best condition of the degree of hydrolysis can be come to 40.40%.The Alcase protease: the ratio of enezyme and substrate:1.4ml,pH:6.5,the temperature of the reaction:55℃,the substrate concentration:10%,the hydrolysis time6h. The best condition of the degree of hydrolysis can be come to47.02 %.The Neutrase protease: the ratio of enezyme and substrate:4.5mL,pH:4.5,the temperature of the reaction:40℃,the substrate concentration:4%,the hydrolysis time4h.The best condition of the degree of hydrolysis can be come to 41.52%.The Flourzyme protease: the ratio of enezyme and substrate: 1.3 mL,pH: 5.5,the temperature of the reaction:50℃,the substrate concentration:11%,the hydrolysis time4h. The best condition of the degree of hydrolysis can be come to 48.79%.The thesis had analysed dissolved free amino acids and the bitter value.
