Safflower,Chinese traditional medica for invigorating the circulation of blood and reducingstasis,is the dried flower peals of carthamus tinctorius L,in which carthamusisone of the most important components.The extraction,purification, structure,the effects of red safflower pigment on antioxidation and hyperlipidemia was systemically studied in the thesis.The results were as follows:With the content of carthamin as an index,the orthogonal design of three factors and three levels were used to select the optimum extraction process.The optimum extracting condition was to add 40 times amount of water,soaking and extracting for 2h every time,2 times totally at 60℃.The extraction rate of red safflower pigment is 78.75%.X-5 resins and the Sephadex LH-20 column chromatograph,a new method on the preparation of carthamin was determined.This method is simple and stable.Until now it has not been reported.Carthamin of purity index isolated by use of the above method,acorrding to its spectrometric data(MS),The molecular weight of the compounds is the same as the molecular weight of cathamin.The antioxidant activity of carthamin extracted and purified from safflower was evaluated by determining the The·O2-,H2O2,and·OH.The results indicated that carthamin could be used as a natural antioxidant in food,cosmetics and medicine Industries.Through the establishment of hyperlipidemia mouse model to study the red safflower pigment effect on hyperlipidemia and antioxidant activity.It was found that the red safflower pigment significantly reduce serum cholesterol(TC),triglyceride (TG),and low-density lipoprotein cholesterol(LDL-C) and at same time markedly increase high density lipoprotein cholesterol(HDL-C) after 6 weeks treatment in hyperlipidemic mice.The result indicated that there were substantial hypolipidemic effects.We further evaluated antioxidant capability.The activities of SOD,ALT,AST and the content of MDA,total antioxidant capacity(TAC) were determined.The results showed that red safflower pigment possessed antioxidant activity and had a protective effect on liver in mice.
